The Study On Effect And Mechanism Of Male Animals' Reproductive System By LBP

Lycium barbarum polysaccharides (LBP),as an important functional factor of Fructus Lycii, has many biologic functions such as adjusting immunity, anti-fatigue and so on. LBP is also used as the strengthening and nourishing medicine in traditional Chinese medicine. However, there isn't literature to report whether LBP has the function of reproductive health or not. This paper discussed the effect and possible mechanism of LBP on male animals' reproductive function and offered the basic information to the field of man reproductive health, which has important meaning on theory as well as application value.To investigate the protective effect and the possible mechanism of LBP on rats' reproductive system damaged by warm bath. 36 Wistar male rats were divided into 6 groups according to their different doses of LBP(Octrl,Onegative Ctrl, 10,50,100,200mg.kg 1d-1) with 6 rats in each. Trauma model of testicle was constructed by warm bath and superoxide dismutase ( SOD ) activity, malonic aldehyde (MDA) contents and morphological change were determined after fourteen days. The results showed testis SOD activity in negative control group were significantly lower than LBP group,and testis MDA content in negative control group were significantly higher than LBP group. The optimal dose is LBP 10mg.kg-1d-1.LBP could protect the spermatogenic cells and promote the normal development of testicle germ cells. LBP can protect rat function of reproductive system whose mechanism may be the anti-oxidation effect and the adjustment of the hypothalamus -pituitary-gonadal axis.To investigate the protective action of LBP on testicle cells' DNA oxidative damage which was induced by hydrogen peroxide (H202). The single cell gel electrophoresis(SCGE) was used to detect the breakage of DNA strand and analyze LBP protection against oxidation damage in testicle cells which were treated by different concentrations of LBP for 1h firstly, and then cultured with 100umol/LH2O2 for 25 min. A certain moment action bycould induce the breakage of DNA strand. The pretreatment of LBP (50 100 200400g/ml) significantly decreased the frequencies of cells with tail moment and the tail length of testicle cells treated by H2O2 LBP itself can not cause the oxidation damage, while it can clean out the free-radical and restrain the testicle cells' DNA damage caused by the oxidative stress.To observe the effects of LBP on sexual function and reproductive function of the hemicastrated rats. 46 male rats were randomly devided into 4 groups.The right testis was extirpated except for normal control group. Normal control group and negative control group were ig given saline; positive control group were hypodermic of testosterone propionate at dose of 2 mg.kg-1; LBP control group were ig given LBP at dose of 10 mg-kg 1d1.Mating tested and erective test was observed. LBP experiment group and positive contrast group can shorten incubation period of penis erects and mounting ; Improve the percentage of mounting animals; The organ coefficient of foreskin gland,seminal vesicle-prostate gland were increased significantly as well as sperm count and vigor . LBP can increase the sexual function and reproductive function of hemicastrated rats. The effective composition was not as substituent of androgen but adjustment of the hypothalamus -pituitary-gonadal axis.To investigate the effect of LBP on sexual hormone of damaged reproductive system of male animals and explore the mechanism of health care function of LBP on reproductive system tentatively. Trauma model of testicle was constructed by warm bath and half castration respectively. Different doses of LBP were paid in front of warm bath (prevention) and after half castration (treatment). The levels of serum T, LSH, FSH, 2 were detected with the Radioimmunoassay (RIA). The levels of serum sex hormone T , LSH , FSH in LBP 10mgkgd group were improved obviously compared with that of the negative control group (P<0.01).
