Objective Taking the NF-?B signal pathway as the entry point,to study the mechanism of reproductive damage of tripterygium wilfordii glycosides?GTW?and lycium barbarum polysaccharide?LBP?antagonizing this damage.It provides laboratory data support for clinical safe use of tripterygium wilfordii glycosides.Methods 40 SPF male SD rats were divided into 4 groups by random number table method,which were blank control group?given 0.5%sodium carboxymethyl cellulose?,tripterygium wilfordii glycosides low-dose Group(given GTW 6.3mg·kg-1·d-1),tripterygium wilfordii glycosides high-dose group(given GTW 12.6mg·kg-1·d-1)and lycium barbarum polysaccharide+tripterygium wilfordii glycosides group(given GTW 12.6mg·kg-1·d-1+LBP160mg·kg-1·d-1),each group consists of 10 rats.Intragastric administration was performed once a day for 8 weeks.After the last gavage,anesthesia,weighing,and dissection of the testis were used to monitor the indicators,which include calculating the organ index after wet testicular weighing;hematoxylin-eosin?HE?staining to observe the pathological changes of testicular tissue;Western bloting to detect the contents of IKB?,p65,p-p65 and Bcl-2 protein.Results1.The general situation:There was no death in each group of rats.The rats in the blank control group grew rapidly and developed well.Mainly manifested as agile response to external stimuli,sensitive action,radiant eyes with no blood congestion,soft and dense fur,no hair loss,good appetite and defecation.Rats in the lycium barbarum polysaccharide+tripterygium wilfordii glycosides group gained good weight,and body weight,testicular mass,and organ index were not different from those in the blank control group.Rats of tripterygium wilfordii glycosides low-dose group and tripterygium wilfordii glycosides high-dose group lost weight to varying degrees compared with the blank group,with slow movements,atrophy,dull red eyes,and dry yellow back hair with epilation,loss of appetite.The stools were loose or even slimy over time,and the testicular quality and organ index were lower than those of the blank control group,with significant differences?P<0.01 or P<0.05?.2.Testicular histopathological results:In the testicular tissue of blank control group SD rats,the seminiferous seminiferous tubules were full,the seminiferous tubules were tightly spaced,the intraluminal cortex was intact,the interstitial cells were arranged neatly and orderly,the numbers of spermatogonia and spermatocytes were normal;In testicular tissue of tripterygium wilfordii glycosides low-dose Group,the seminiferous tubules became edema and twisted,the seminiferous tubules became wider,the spermatogonia and spermatocytes decreased;In the testis tissue of tripterygium wilfordii glycosides high-dose group,the seminiferous tubules had increased edema,the seminiferous tubules became wider,the lumen endothelium became thin,even loose epithelium appeared,the interstitial cells were disordered,spermatogonia and spermatocytes significantly reduced;Compared with the blank control group,the lycium barbarum polysaccharide+tripterygium wilfordii glycosides group showed no significant difference in the shape of seminiferous seminiferous tubules,the seminiferous seminiferous tubules had a wider interval,but the amplitude was lower than that of other GTW groups.The number of spermatogonia and spermatocytes decreased slightly,and rehearsal of mesenchymal cells was acceptable.3.Expression results of IKB?,p65,p-p65,and Bcl-2 proteins in testis tissues:Compared with the blank control group,the expression levels of p65 and p-p65 protein in tripterygium wilfordii glycosides low-dose and high-dose groups were significantly increased?P<0.01?,and IKB?and Bcl-2 protein levels were significantly reduced?P<0.01?,the expression levels of p65,p-p65,and Bcl-2 in lycium barbarum polysaccharide+tripterygium wilfordii glycosides group were significantly increased?P<0.01?,and IKB?expression level was significantly reduced?P<0.01?;Compared with tripterygium wilfordii glycosides low-dose group,the expression levels of p65 and p-p65 protein in tripterygium wilfordii glycosides high-dose group increased to varying degrees?P<0.01 or P<0.05?,the expression level of Bcl-2 decreased significantly?P<0.01?.There was no significant difference in the expression level of IKB?.The levels of p65,p-p65,IKB?,Bcl-2 protein expression in rats of lycium barbarum polysaccharide+tripterygium wilfordii glycosides group significantly increased?P<0.01?;Compared with rats in tripterygium wilfordii glycosides high-dose group,the expression levels of p65,p-p65,IKB?,and Bcl-2 protein in testicular tissues of rats in lycium barbarum polysaccharide+tripterygium wilfordii glycosides group were significantly increased?P<0.01?.Conclusion1.The degree of reproductive damage of tripterygium wilfordii glycosides on male SD rats is closely related to the dosage.2.Tripterygium wilfordii glycosides may regulate the expression of related proteins in the NF-?B signaling pathway,such as up-regulating p65 and p-p65 protein expression levels,down-regulating IKB?and Bcl-2 protein expression levels,promoting cell apoptosis and exerting its reproductive damage Role.3.Lycium barbarum polysaccharide may increase the expression levels of p65,p-p65and Bcl-2 proteins and down-regulate the expression level of IKB?protein,thereby activating the NF-?B signaling pathway,inhibiting germ cell apoptosis,and antagonizing the reproductive damage of triptolide.
