Periodontitis is usually characterized by inflammatory cells accumulation in the exhavascular connective tissue. Adhesion molecules play an important role in the pathogenesis of periodontal disease by promoting the recruitment and retention of leukocytes in periodontal tissue. In the present studies, we studied the capacity of interleukin ?1?3 (IL條(3)to regulate expression of intercellular adhesion molecule - 1 (ICAM - 1) on clinically healthy human gingival fibroblasts (HGF) and periodontal ligament cells (PDLC) and the effects of Baicalin on IL- 1(3 induced expression of ICAM- 1 in HGF and PDLC. Then we observed expression of ICAM- 1 in the gingival tissue of periodontitis and healthy subjects. Experiment 1 The effects of Baicalin on IL - 1?induced expression of ICAM - 1 in HGF and PDLCThe purpose of this study was to investigate the capucity of IL - 1(3 to regulate intercellular adhesion molecule - 1 expression on clinically healthy HGF and PDLC, and then we observed the influence of Baicalin on the IL - 1(3 induced experession of ICAM - 1 in HGF and PDLC. In the experiment, Immunohistochemical S-P method was used.The levels of ICAM -1 expression in IL - 1(3 treated HGF and PDL fibroblasts were statistically significant (p<0. 01) compared to normal untreated controls. The levels of ICAM- 1 expression in Baicalin (lOng/ ml.100ng/ml and 1000ng/ml) treated HGF and PDLC were not statistically different from controls. Addition of HGF (1000 ng/ml ) and IL- 1(3 (Ing/ml) could decrease ICAM- 1 expression.This study demonstrated that IL - 1(3 upregulated ICAM - 1 expression in both HGF and PDLC. Baicalin can sppress ICAM - 1 expression on HGF and PDLC. This result indicated that ICAM - 1 was involved in the pathogenesis of periodontal disease, which provoded an important adhesion pathway for leukocytes migration into gingival sulcus in periodontitis lesions. Baicalin can decrease ICAM- 1 expression on HGF and PDLC, and inhibit the adhesion of leukocyles and fiberblasts.Experiment 2 The study of expression of ICAM - 1 in gingival tissueIn order to reveal the differences of expression of adhesion molecule between health gingival and periodontitis tissue, we have facused on the expression of ICAM ?1 in gingival tissue of health subjects and periodontitis.The expression of ICAM - 1 in the gingival tissue of periodontitis and health subjects were studied by immunohistochemical S - P method. ICAM ?1 positive leukocyles were observed in the gingival connective tissue of periodontitis, which had statistically significant differences between periodontitis and health subjects. .Functional epithelium and apical partiod sulcus epithelium expressed ICAM - 1 in both healthy and periodontal tissue. This result indicated that ICAM - 1 expression in the gingival tissue of periodontitis was more than that in health subjects, which may provide an important adhesion pathway for leukocytes migration into gingival sulcus in periodontitis lesions. The overexpression ofICAM ?1 in gingiva was relevant to the retention of inflammalory cells in periodontitis lesions and increased the local destruction of periodontal tissue.
