LPS is one of the most important factors in the causes of peridental disease. It can cause the damages of peridental tissues and loss of teeth. PDLC is a key part of periodontal tissues and it can differentiate into different cells and keep healthy of periodontal tissues. Researches have found that LPS can damage PDLC directly. It can inhibit the activities of the cells, ALP and damage the ultrastuctures of the cells. Gallnut water extract is mainly made up of tannin; it has many characters, such as anti-bactierier. Gallnut water extract is also found to play an important role in the prevention and treament of dental caries and periodontal disease. We have observed that gallnut water extract has influence on the decreaseing of activities of PDLC, damageing of the cell ultrastructures, decreasing the attachment on the slices of cement and decreaseing the activities of ALP caused by LPS. The results prove that gallnut water extract can protect PDLC damaged by LPS.1. The effects of gallnut water extract on the activities of PDLC inhibited by LPSThe cell culture technique,3 H-TdR and transmission electron microscope (TEM) methods are used to observe the effects of the gallnut water extract on the activities of PDLC inhibited by LPS. The results indicate that at the concentration of 100g/ml, LPS can inhibit the activities of PDLC remarkably and PDLC' activities can greatly improved after adding gallnut water extract. Gallnut water extract is divided into two groups. One group adds gallnut water extract before LPS(A group) and the other group adds them at the same time (B group). And both of them can improve the activities of PDLC inhibited by LPS, especially the former. So we conclude that gallnut water extract has protective effects on PDLC activities inhibited by LPS.2. The effects of gallnut water extract on the ultrustructure of PDLC damaged by LPSThe cell culture technique and transmission electron microscope (TEM) methods are used to observe the effects of gallnut water extract on the ultrastructures of PDLC damaged by LPS. The result indicate that LPS group has significant impairmental effects on PDLC' ultrastructures in 100g/ml, especially to the mitochondria. The impairment of ultrastructures of PDLC can greatly improve after adding gallnut water extract. Gallnut water extract is divided into A group and B group, and both of them can relieve the impairment of ultrastructures of PDLC caused by LPS, especially the former. The conclusion is that gallnut water extract has protective effects on PDLC' ultrastructures damaged by LPS.3. The effects of gallnut water extract on the attachment of PDLC inhibitedby LPS on the slices of cementTo observe the effects of gallnut water extract on the attachment of human PDLC inhibited by LPS on the slices of cement, we use the cell culture technique, cell counting and scanning electron microscopy (SEM) .We find that contrasting to the control group, LPS group has inhibitory effects on PDLC' attachment on the slices of cement in vitro in 100g/ml. The results of cell counting and SEM show that the number of cells attached to root slices deceases and the extension of cells becomes inadequate. The effects can greatly improved after adding gallnut water extract. Gallnut water extract is divided into A group and B group and both ofthem can increase PDLC' attachment on the slices of cement in vitro, especially the former. So we draw a conclusion that gallnut water extract has protective effects on the attachment of human PDLC on the slices of cement inhibited by LPS. 4. The effects of gallnut water extract on the activities of ALP inhibited by LPSWe use the cell culture technique and enzyme kinetics methods to investigate the effects of gallnut water extract on the activities of ALP of PDLC inhibited by LPS. The results indicate that at the concentration of 100 ug/ml, LPS can inhibit the activities of ALP.
