Articular cartilage defect caused by all kinds of reason is always important in joint surgery, particular in big square articular defect. Artificial catilage by the method of tissue engineer is necessary trend to repair cartilage defect. However; when it comes to cartilage tissue engineer material, particular in artificial synthesizing high molecule material, question arise, the question is these material having bad quality such as low hydrophile, low cell absorbing and that cell cannot easily take interaction with material,, hence; the key is how to improve the cells' absorbance on material;how to increase cells and function expression, the object of this expencement is that The effect of collagen IIwhich is natural absorbing material on the adhesion and differentiation of induced mesenchymal stem cells(MSCs)was evaluated on PLGA which is a artificially absorbing material.Materials and MethodsThe 4-8 week-old female or male the New Zealand rabbit offered by the experimental animal center of Henan province were used in study. The autologous mesenchymal cells derived from the midshafts offemur bone marrow of New Zealand Rabbits were havested. With insulin-like growth factor I (IGF- I ) and transforming growth factor beta 3 (TGF-P 3) , the cells were induced and expanded in cell culture. MSCs obtained from induced two weeks were culture on PLGA coated by collagen II in F-12 containing 10% bovine serum, while culture on the simple PLGA as control, checking up the conditions of growth of the cells by applying some Methods:1 by using RT-PCR to obersve whether MSCs do defferential to chondrocyte, whether MSCs which was induced express collagen II which is only emerge in chondrocyte product.2 cells counting can estimate cell adhesion .3 immunohistochemistry can estimate collagen II expressing of MSCs which is induced on experimental group (PLGA coated by collagen II) and control group (the simple PLGA) .4 and MTT test can estimate cell growing and estimate cell condition of growth , adhesion, and differentiation.5 The data were analyzed by software SPSS10. 0. by using the X2-test or Fisher's exact test and GLM-Univariate Measure). A two-side P valueof less than 0.05 was considered statistically significant. Result:1 experimental group (culture medium having growth factor) express procollagenllmRNA, on the contrary , the control groups (culture medium not having growth factor) donot express procollagenllmRNA, so procollagen IlmRNA in the each groups had singnificant differences.2 after cell tansplanting 2, 4, 6, 8 hour each group cell number was treated by stastic and show: experiment group and control group had singnificant differences (P<0. 01) in 6, Shour.3 immunohistochemistry was applying to check collagenII of iMSCs which was induced on each groups. The PLGA groups express low, whereas the PLGA coated collagen II group still remain strong postive on 30 days. Experiment group and control group had singnificant differences (P<0. 05) .4 MTT method which was using to estimate the cell alive, cell proliferation in different groups shows that cells was singnificant lower in proliferation in PLGA group than in PLGA coated collagen II group, experiment group and control group had significant differences (P<0. 01) in 1, 2, 3 week.Conclusion:the result of experiment show:1 MSCs which was induced can transfer to deferentiation of chondrocyte, and can also express collagen II which is only emerge in chondrocyte product.2 the PLGA coated collagenII group can make MSCs adhesion fasten on the surface and also increase the cells proliferation;and continue expressing collagenII which is only emerge in chondrocyte product. So the material of the PLGA coated collagenII can provide three dimension solid structure which benefit the cell alive.
