Objective: To study the protective effect for Chinese medicine of promoting blood circulation and removing blood stasis on vascular endothelial function, aimingatfurther research on the pathogenesis of atherosclerosis, and to seek for new thinking and effective drugs in protecting vascular endothelial function.Methods: SD rats were randomly divided into five groups: control group (CG), model group (MG), isosorbide dimitrate group (IDG), small dose salvia miltiorrhizae group (SMS), high dose salvia miltiorrhizae group (SMH). Establishing the models of atherosclerosis by feeding hyperlipidemic feeding to the rats for four weeks, then remeasuring the blood rheology, and the levels of serum total cholesterol (TC) and triglyceride (TG). The treated groups have been taken different dose salvia miltiorrhizae, isosorbide dimitrate,et al for four weeks. Before the end of the experiment, establishing myocardial ischemic rat models of blood stasis with endothelial-cell injury by means of giving rats an injection of hydrochloride adrenalin (Adr), pituitrin (Pit) and making them swim in ice-cold water for five minutes. Writing up the change of SD rats' weigh and electrocardiogram (ECG) in different times. The levels of plasma endothelin-1 (ET-1), serum nitric oxide (NO) and nitric oxide synthase (NOS) were assayed by radioimmunoassay method, enzyme-linked immunoassay and biochemical techniques. We also studied the effect of different dose salvia miltiorrhizae on the metabolism of nitric oxide and endothelin-1. Results: The concentration of plasma ET was significantly higher in model group (140.82±25.71pg/ml) than in control group (81.53±15.44pg/ml, P< 0.01). Compared with the model group, small dose salvia miltiorrhizae group and isosorbide dimitrate group significantly increased the level of ET (90.34±15.19pg/ml, 92.53±11.74pg/ml, P<0.01,).ET from high dose salvia miltiorrhizae group(126.70±13.44pg/ml)had high tendency but not reached statistical difference(P>0.05).It is suggested that salvia miltiorrhizae could partly reduce the levels of plasma ET. The salvia miltiorrhizae was able to increase the content of serum NO. The concentration of serum NO in the model was 12.97±3.25umol/L, the difference was significant in comparison with those in control group (26.17±6.44umol/L, P<0.01). The levels of serum NO in the three treatment groups were remarkably higher than in the model group(SMS 25.98±6.30umol/L,SMH20.43±5.40umol/L,IDG20.08±3.30umol/L, P<0.01~0.05). The concentration of serum TNOS in SMS group and in SMH group were 22.99±3.26umol/L, 21.33±2.08umol/L,significant difference were shown in comparison in with those in the model group(14.65±1.26umol/L ,P<0.01). The level of iNOS in SMS group ( 12.95±2.11umol/L) was increased ,compared with the model group(6.63±1.09umol/L), there was significant difference(P<0.01), The level of iNOS was significantly higher in SMS group than in ID group(11.08±2.99uoml/L,P< 0.05), but the level of iNOS in SMH group (8.93±0.69umol/L) showed insignificant difference(P >0.05). The salvia miltiorrhizae was able to decrease the content of serum total cholesterol (TC) and triglyceride (TG), compared with the model group, there was significant difference (P<0.05), while compared with the ID group, there was insignificant difference (P >0.05). It is suggested that the function of decreasing the content of serum total cholesterol (TC) and triglyceride (TG) by salvia miltiorrhizae was less superior than isosorbide dimitrate. Salvia miltiorrhizae was able to inhibit the whole blood viscosity and the plasma viscosity, and blood rheology was improved.Conclusions: 1Cardiovascular risk factors contribute to atherogensis by inducing endothelial-cell injury and dysfunction. The endothelial dysfunction may be promoted the progression of cardiovascular disease, and affect the synthesis, release of vasomotory material ET and NO. So the level of ET may be taken as the supplemental indexes for evaluating the numbers and severity of atherogenesis, and to seek for new thinki...
