Objective: Serum paraoxonase-1 (PON-1) is a Ca2+ - dependent arylesterase enzyme that is associated with high-density lipoprotein and has been shown to protect low density lipoprotein from oxidation and to remove lipidperoxidation products. Two common polymorphisms of the paraoxonase gene a Gln to Arg interchange at position 192 and Met to Leu interchange at position 55 have been demonstrated to modulate serum PON-1 activity toward paraoxon, a nonphysiological substrate. Leu 55 (allele L) and Arg 192 (allele B) are associated with higher activity than Met 55 (allele M) and Gln 192 (allele A), respectively. Previous studies have not agreed whether two polymorphisms of PON-1 are genetic risk factors of coronary heart disease (CHD). The present study was designed in a sample of Chinese subjects to investigate the correlation of codon 55, 192 gene polymorphisms of PON-1 and coronary heart disease and its severity of stenosis.Methods: 151 patients with CHD (according to the undergoing diagnostic Judkins selection coronary angiography ) and 61 subjects without CHD (according to the undergoing diagnostic Judkins selection coronary angiography) were enrolled . The genotype and allele frequency of PON-1 codon 55, 192 gene polymorphisms were assayed by Polymerase Chain Reaction (PCR)-restriction fragment length polymorphisms (RFLP) technical. The coronary score of Gensini was estimated the severity of the coronary artery stenosis. Statistical analysis: 1.The basic characteristics between the CHD group and the control group was analyzed first .All the numerical date was denoted by chi-square (χ2) test, the measurement date who are normal distribution and homoscedasticity was denoted by Student's t test,the other date was denoted by rank sum test by Wilcoxon(Size of a testα=0.05).2. The frequencies of genotype and allele were compared between with and without CHD group using the χ2 test; The frequencies of allele were calculated by Hardy-Weinberg equilibrium model; The distribution of genotype frequencies between sample and population was compared to the Hardy-Weinberg equilibrium model using the χ2 goodness-of-fit test; Fisher's exact test was performed to compare allele frequencies between these groups (Size of a testα=0.05). 3. The relationship between genotype of PON-1 gene codon 192 and Gensini's coronary score in CHD subgroup was compared by Kruskal-Wallis H test (A two-tailed value of probability≤0.05 was accepted as statistically significant).All calculation were performed using the SPSS 11.5 program for windows .Results: 1. The basic characteristics of study population: The percentage of sex and smoking history in the CHD group and the control group conformed to normal distribution by χ2 test , the value of χ2 is 10.382 and 4.925 , the value of P is 0.001 and 0.035 , respectively . There was significance difierence between the two groups (Size of a testα=0.05). The percentage of male and smoking in the CHD group were higher. The Student's t test were applied to those value of variable, such as age , SBP , DBP , BMI , TG , TC , apoA , apoB , apoA/apoB , the value of t were 1.778 , 1.726 , 1.866 , 2.366 , 2.110, 0.301 , 1.963 , 0.462 , 1.838 , the value of P were 0.077 , 0.087 , 0.061 , 0.019 , 0.036 , 0.746 , 0.051, 0.644 , 0.068 , respectively . It is believed that there was difference between the two groups about BMI and TG (Size of a testα=0.05) . The value of GLU , HDL-c and LDL-c was denoted by Wilcoxon rank sum test . The value of u were 0.846 , 1.767 and 0.496 , the value pf P is 0.398 , 0.077 and o.620 , respectively . So we can say there was no difference between the two groups (Size of a testα=0.05) .2. Relationship between the sample and population of PON-1 codon 55 , 192 genotype distribution : We found that PON-1 gene codon 55 with restriction enzyme digestion has three genotypes : MM , ML , LL , and PON-1 gene codon 192 with restriction enzyme digestion has three genotypes too : AA , AB , BB, in this study population .
