| Objective: Evaluated the incidence of liver metastasis in normal liver and cirrhosis liver by animal experiments and clinical observation, to reveal the risistance of cirrhosis to metastatic cells and it's possible mechanisms.Methods: 1,Developed the mice model of liver cirrhosis by gastro-gavage with carbon tetrachloride and developed the liver metastases model by intrasplenic injection of colonic adenocarcinoma cells (colon26). Observed the incidence of liver metastasis in liver cirrhosis mice and mormal liver mice inoculated with colon26. Examined the expression of Liminin(LN),Fibronectin(FN) and E-selectin in normal liver group("A1" group), cirrhosis liver group("B1" group),normal liver inoculated with colon26 group("A2" group) and cirrhosis liver inoculated with colon26 group ("B2" group) by immunohistochemistry(IH). 2,435 patients with colorectal cancer were divided into two groups: normal liver group and cirrhosis liver group, compared the incidence of liver metastasis in two groups. Results: 1 At the first stage of "A2" group after being inoculated with colon26, there were no obvious changes in spirit, activity, diet and body configuration. One week later, mice became thin, their activities were slower than before and spirit became worse. Drinking and eating reduced obviously. Major mice inoculated developed liver metastasis at the 10th day postinjection, there were lots of metastasis mass in different size in every lobe of liver. Other organic metastasis were not observed. There were gigantic metastasis mass in a part of mice and hemorrhagic ascites in the abdominal cavities. Grey metastasis mass only can be seen in the site of spleen inoculated with colon26 in individual mice and liver metastasis were not observed. There were no obvious changes in spirit, activity , diet and body configuration in major mice of"B2"group after being inoculated with colon26. A week later, a few mice became low spirit and thin. Their drinking and eating reduced. On the 10th day after being inoculated, there were small cirrhosis tubercles full of liver and matastasis mass were not observed in most mice. Grey and small matastasis mass scattered in a few mice. The incidence of liver metastasis in "B2" group(4 of 16, 25%) was significant lower(P= 0.001) than that of "A2" group(14 of 16, 87.5%). 2 The expression of Liminin,Fibronectin and E-selectin in the four groups. â‘ The expression of LN. Light microscopically examinations showed that in the liver tissues of "A1" group and "A2" group, there was a little LN and most of them distributed in portal area, minority in hepatic lobules, taking the form of filaments or particles, light yellow to brown yellow. There was much LN in "B1" group and "B2" group distributed in portal area, hepatic lobules and fibrous tissue, taking the form of filaments or particles, brown yellow to brown. The expression of LN in "B1" group was significantly higher than that in "A1" group (P<0.0001). The expression of LN in "B2" group was significantly higher than that in "A2" group (P<0.0001). There was no significant difference in expression of LN between "A1" group and "A2" group, "B1" group and "B2" group(P>0.05). â‘¡ The expression of FN. In the liver tissues of "A1" group and "A2" group, there was a little FN and most of them distributed along the wall of hepatic sinusoid and in the matrix of portal area, light yellow to brown yellow. In "B1" group and "B2" group, the expression of FN can be seen in the wall of hepatic sinusoid, portal area and fibrous tissue, brown yellow to brown. The expression of FN in "B1" group was significantly higher than that in "A1" group (P=0.001). The expression of FN in "B2" group was significantly higher than that in "A2" group(P<0.0001). There was no significant difference in expression of LN between "A1" group and "A2" group, "B1" group and "B2" group(P>0.05). â‘¢ The expression of E-selectin.
