Objective To study the difference of gene expression in hepatocellular carcinoma(HCC) exhibiting different invasion and metastasis. Methods By using pairwise combination of one fluorescent anchored primer and one arbitrary primer, the protocol for fluorescent differential display-PCR (fluoro DD-PCR) was developed .Then using the developed fluoro DD-PCR techniques, we isolated several differentially displayed fragments .After analyzing the candidate fragments by means of RNA blot ,we identified and sequenced the true clone, and then used BLASTn to search for the homology in GenBank. With using immunohistochemistry, the differentially expression of the gene was tested in 45 HCCs.Results We fond 3 (S1-3) candidate fragments , but only one fragment(S2) was identified as true positive ,and the sequencing analysis of S2 revealed that had high homology with known sequence of the human Matrix metalloproteinases 1(MMP-1) gene . By analyzed with immunohistochemistry in 45 HCCs ,the MMP-1 expression in HCCs with invasion or metastasis is higher than that without invasion and metastasis (P<0.01),but there was no significant difference between the later and the no-carcinoma tissues(P>0.05). Conclusion The high-level expression of MMP-1 in HCC may be related to the of the tumor.
