AIM: To isolate, purify the polysaccharides from Aloe barbadensis Miller, assay the soluble sugar content, analysis the composition and structure, and study the antitumor effect on mice in vivo.METHODS: The polysaccharides of Aloe barbadensis Miller gel was precipitated with ethanol and further purified by chromatography in a Sephadex G-100 column. Its total soluble sugar content was detected colorimetrically by the anthrone-sulfuric acid method with manose as standard. Gas chromatography, ultrared spectroscopy, nuclear magenetic resonance, periodate oxidation analysis and Smith degradation were used to study the structure of this polysaccharide. Then demonstrate the antitumor activities of Aloe polysaccharide on mice transplantation tumor S180,U14,H22, and study its potentiation and attenuation on chemotherapeutic agent.RESULTS: A polysaccharide named AP I was isolated from the fresh Aloe barbadensis Miller leaf, AP I was mainly ((1→4)-D-acemannan, the total sugar content was 81.11%. AP I could inhibit implanted carcinoma S180,U14,H22 growth in mice, with cyclophosphamide it could enhance the antitumor effect and lessen the toxic-side effects. CONCLUNSION: AP I (((1→4)-D-acemannan) from the fresh Aloe barbadensis Miller not only antagonized various transplantation tumor, but increase the action of cyclophosphamide and reduced its toxic-side effects.
