Objective:To set up a method for simultaneous determination of Aloin and Aloe-emodin in Aloe by HPLC, and to develop in the preparation of the content standards.Methods:Extract an active constituent by the fact that alcohol extracts law from aloes in powder;Using a column of ZORBAX-SA-C18 (5μm, 200mmX4.6mm)with a mobile phase of methanol-water(70:30), flow rate being on the move:1.0 mL/min, the post temperature:25℃, enters appearance amounts:20μL. Aloin was detected at 360 nm and Aloe-emodin at 430 nm.Results:It was shown that Aloin had a good linear relationship within the range of 0.5~100μg/mL (R=0.99975), and a good precision degree (RSD=0.7%), and the average recovery was 98.32%(RSD=0.4%); Aloe-emodin had a good linear relationship within the range of 0.25 100μg/mL and good precision degree (R=0.9999), and a good precision degree (RSD=1.2%), and the average recovery was 99.36%(RSD=0.41%).Conclusion:HPLC is rapid, simple and accurate, and could be used for the simultaneous determination of Aloin and Aloe-emodin in Aloe at the same time.
