Objective:To investigate the effect of the compound of allograft cancellous bone and selecctive inhibitor of cNOS or (L-NAME ) iNOS (AG)on repairing radius bone defects in rabbit. Methods:To make allograft cancellous bone and compound with selective inhibitor of cNOS L-NAME and selective inhibitor of iNOS AG.The 40 New Zealand white rabbits were divided into two groups. 10mm segmental bone and periosteum defects were created on bilateral radius to make the animal models of bone defect .Group A, the simple compound of cancellous allograft bone were implanted on left limb and the compound of cancellous allograft bone and selective inhibitor of cNOS L-NAME were implanted on right limbï¼›Group B, the simple compound of cancellous allograft bone were implanted on left limb and the compound of cancellous allograft bone and selective inhibitor of iNOS AG were implanted on right limb. The effect were observed by gross,histooathological X-ray examinations and the bone mineral density (BMD) at 2,4,8,12 week after operation. The result of X-ray examinations were evaluated by computer image analysis to calculate callus gray degree. The healing of bone defect was evaluated by the histooathological examination.BMD was measured by dual energy x-ray absorptiometry (DEXA), using a Hologic QDR-1000TM x-ray bone densitometer. defects region (R1) analyses were performed. The amount and speed of new bone formed on region of bone defect were evaluated by BMD. Mechanical strength was measured by three points curving test at 12 weeks after operation and compared with normal radius .Results:The compound of cancellous allograft bone and selective inhibitor of iNOS AG were superior in term of the amount and speed of new bone formed on region of bone defect to the simple cancellous allograft bone. The simple cancellous allograft bone were superior to the compound of cancellous allograft bone and selective inhibitor of cNOS L-NAME. The defect implanting the compound of cancellous allograft bone and selective inhibitor of iNOS got basically healing at 12 weeks. The result of Mechanical strength had no statistically significant difference with normal radius .Conclusion:Inhibitor of NOS had effect on repairing bone defect. Inhibitor of cNOS can inhibit repairing bone defect .Inhibitor of iNOS can improve repairing bone defect.
