Purpose: To study the antiproliferative effects of α-tocopherol acetate on the glaucoma filtering surgery .Methods: Twenty-four pigmented rabbits were employed auto-control. All eyes were randomly divided into experimental group and control group . All rabbits(48eyes) underwent trabeculectomy.Twenty-four hours before surgery ,the treated eyes were injected subconjunctivally with 0.5 ml of 100mg ATA in 0.75% ethanol in balanced saline solution (BSS).The control eyes were injected subconjunctivally with 0.5ml of 0.75% ethanol in BSS. The 2,7,14,21day after surgery ,we observed the intraocular pressure(IOP),filtering bleb and inflammatory reation .PCNA in filtering pathway were detected by using immunohistochemical assay.Results: The 2 , 7 ,14 day after surgery ,Intraocular ptessure (IOP) was significantly lower in treated group (ATA) than in the control group (P<0.05andP<0.01).Light conjunctival congestion and inflammatory in anterior chamber had disappeared 3 days after surgery and haven't the corneal edema and other complications.The number of functioning blebs without connective tissue collagenization were showed statistically significant differences between the control and treated group the 7 and 14 day after the surgery(P<0.01andP<0.05).there were statistically significant differences in the expression level of the PCNA between two groups (P<0.05).Conclusion: ATA have the efficacy of inhibit the fibroblast cells proliferation in filtering pathway,show better IOP control and bleb survival in this experimental model of filtering surgery.
