Matrix Metalloproteinase-1 And Tissue Inhibitor Of Metalloproteinase-1 In Endometriosis

Objective:To explore the level of matrix metalloproteinase-1(MMP-1) and tissue inhibitor of metalloproteinase-1(TIMP-1)in peritoneal fluid(PF)and serum and their etiologic roles in endometriosis(EMS),and to study the clinical significance of serum TIMP-1。Methods:PF and serum was obtained from patients with endometriosis(n=44,8 women with Stage I,2 women with StageⅡ,20 women with StageⅢ,14 women with Stage Ⅳ)and from women without endometriosis(n=30)as controls,and some serum was obtained from EMs patients(n=18)followed by pharmacotherapy after laparoscopy,all samples were analyzed for MMP-1,TIMP-1 by Enzyme Linked Immunosorbent Assay(ELISA)。Result:1,The MMP-1 concentrations in PF and serum were significantly higher in patients with endometriosis compared with those of the controls(P<0.05),while the TIMP-1 concentration in PF and serum were lower than the controls(P<0.01)。After clearance of endometriosis foci with laparoscopy and pharmacotherapy in three month,the serum TIMP-1 concentration increased significantly(P<0.01)。2,The women without endometriosis,it has linear negative correlation relationship between PF MMP-1 and PF TIMP-1;while in patients with endometriosis,there has no correlation relationship between them。It had a linear negative correlation between the EMs RAFS marks and TIMP-1 level in PF and serum。 3,Suppose the normal range of Serum TIMP-1≤335ng/ml,and the sensitivity of EMs was 75%,specificity was 88%。Conclusion:The high level of MMP-1 and the low level of TIMP-1 in peritoneal fluid and serum of patients with endometriosis may play an important role in pathogenesis and development of the disease,and serum TIMP-1 may have important value in diagnosis,postoperative follow-up,surveillance and evaluation of the effectiveness of surgery and drug therapy.
