Abstract/Summary:
Aspergillus is an ubiquitous fungus around the world, and members of the genus cause a group of diseases known as aspergillosis. Aspergillus versilocor is a common environmental fungus. The allergenic bioareols such as spores, mecelium or fragments of this genus have great relationship with the aspiratory symptoms that may be in the form of invasive infection, colonization, toxicoses or allergy. The purification, identification of antigens and allergens of Aspergillus versicolor have not been reported so far. In present study, more than 20 kinds of allergen proteins have been separated from the crude extract of the fungus by SDS-polyacrylamide gel electrophoresis(SDS-PAGE);3 major allergens with molecular weights of 17kDa, 20kDa and 67kDa, respectively, and 4 minor allergens with molecular weights of 19kDa, 22kDa, 35kDa and 76kDa, respectively, have been identified by Western-Blotting with the antisera of Aspergillus allergic patients. After being eluted through DEAE-cellulose (DE-52) chromatography and Sephacryl S-200 gel filtration, the allergens composing of the major allergen with molecular weight of 20kDa and the minor allergen with molecular weight of 35kDa were purified and then identified by Western-Blotting. The recovery of the proteins is 31.84%.
