To elucidate the radiation effect on the proteome of rat peripheral blood lymphocytes, the proteome levels before and after ionizing radiate were compared by the technic of two-dimensional electrophoresis (2-DE). The proteir levels were measured at different times of 24h and 72h after the 2Gy total-body irradiation. The effect of different doses were also analysed for 1, 2, 4 and 6 Gy total-body irradiation. The total protein samples prepared by one-step method were separated by the h: gh resolution 2-DE and silver stain, and the resulted spots were detected by the Imagemaster 2DE software.Seven protein spots disappeared and sixteen protein spots showed significant differences in quantity at 24h and 72h after the 2Gy total-body irradiation. In the 1, 2, 4 and 6Gy irradiation groups, two protein spots disappeared, Two other spots was up-regulated and thirteen spots were down-regulated vdth increase of the irradiation doses. The results showd reveals that ionizing radiation could induce significant changes in protein expression of rat peripheral blood lymphocytes. The time-effect and dose-effect relations could be induced for the expression of some protein spots upon irradiation, which might be a reflection of the response to the irradiation damages.
