Immunoprotective effect of HSH resuscitation on the rat with hemorrhagic shock Abstract Objective:To evaluate the protective effects of HSH resuscitation on the immuefunction of rat with hemorrhagic shock Methods: Controlled hemorrhagic shock was induced by blood withdrawal(mean arterial pressure, 40 mm Hg) and maintained for60 min. The animalswere randomly divided into three groups:group 1 (n = 6) was sham-operated,group 2 (n = 18) was treated with RL, and group 3 was treated with 6 mL/kgHSH(n = 18). Group 2,3subdivided 3 subgroup ,The rats were killed 1,2,4h after resuscitation. HE staining,Transmission Electron Microscopy,TUNEL staining were used to detect the apoptosis of lymphocyte in spleen ,Apoptosis index (AI) of each group was calculated. Meanwhile realtimequantitive RT-PCR was performed to record the TNF-α mRNA copies inspleen. Results: Billroth's cord widen under lightscopy in RL group,in 4h subgroupdot necrosis developed . only Billroth's cord widen in HSH group,no suchchanges occurred in sham group.Typical Morphological changes such aschromatin becomes pyknotic and condensed into sharply delineated masses present atthe edge of the nuclear envelope, cell volume decreases, cytoplasmic organellescompact, and cell density increases.occurred.In TUNEL staining a small quantity ofpositive cells located in germinal center of lymph node in PALS district ,noapoptotic bodies found .apoptotic cells increased in 1h subgroup of RL group , alsoapoptotic cells was found in red pulp.Apoptosis top in 2h subgroup of RL group ,apoptotic cells widespreaded in white pulp and red pulp,these become silght in 4hsubgroup.Apoptotic cells dot in PALS district in all HSH group. The AI of HSH and1h subgroup of RL had no difference of sham group(p>0.05).The 2,4 h subgroup ofRL group was higher than other groups (P<0.05) , no differents in HSH第二军医大学硕士学位论文 英文摘要 第5 页subgroup(p>0.05).The expression intensity of TNF- α mRNA was higher inRL(except 4h subgroup)and HSH group than sham group (P<0.05),1h subgroup ofRLgroup higher than 2h,4h subgroup and 1h,2h subgroup of HSH. Conclusion: Use of HSH to resuscitate hemorrhagic shock can downregulatethe expression of TNF-αmRNA in spleen ,reduce the excessive apoptosis oflympocytes in spleen,and protect the immunofunction following hemorrhagic shock .
