The Expression Of TNF-α And IL-1β MRNA In Lung Tissue After Liver Ischemia-Reperfusion In Rats

OBJECTIVE To observe acute lung injury induced by liver ischemia/reperfusion in Wistar rats and the expression of tumor necrosis factor- α and interleukin-1β mRNA in liver and lung tissue, and study its mechanism.METHODS A model of liver ischemia-reperfusion injury was established by imitation.48 male Wistar rats were randomly divided into 6 groups as follows: the control group (the group of sham operation), simply ischemia 30 min without reperfusion (I group), reperfusion following ischemia 30 min (I/R group), 1 hr reperfusion following ischemia 30 min (I/R1h group), 2 hrs reperfusion following ischemia 30 min (I/R2h group) and 4 hrs reperfusion following ischemia 30 min (I/R4h group). TNF- a was detected by immunohistochemical staining and IL-1β mRNA was defined by site hybridization technique in lung and liver tissue.GPT,AKRand Y -GT in plasma were also measured .RESULTS Histological appearance in liver was inflammation cell infiltrated,degeneration and local necrosis were found .In lung, there were the inflammatory infiltration of the neutrophilic granulocyte, blood capillary hyperemia and pulmonary alveolus interstitial became thicken.The expression of TNF-α and IL-1β mRNA were significantly increased.especially in ischemia group and after reperfusion 1,2h group(P<0.01;P<0.05). GPT AKP and γ -GT have significant higher in ischemia group and ischemia/reperfusion group(P<0.01,P<0.05).CONCLUSIONS These results suggest that acute lung injury happened after liver ischemia/reperfusion. TNF-α and IL-1β may be play an important role in this process,two factors expressed in early phase after reperfusion.
