Objective:To study the Pae and Oxy direct effect on the degranulation function of RBL-2H3 mast cells.Methods: 1. Different concentration of Pae and Oxy influenced the proliferation of RBL-2H3 mast cells determined by MTT colorimetric assay. 2. Established a pharmic model on the degranulation of RBL-2H3 mast cells (rat basophile leukemia cell line), activated by compound 48/80(10 μg/ml). The histamine and tumor necrosis factor-alpha(TNF-α) from the supernatants, the spontaneously released histamine and the total histamine were all determined by ELISA. 3. Pretreated RBL-2H3 cells with different dilutions of Pae and Oxy, and activivated by compound 48/80(10 μg/ml).The histamine and tumor necrosis factor-alpha(TNF-α) from the supernatants, the spontaneously released histamine and the total histamine were all determined by ELISA. The percentage of histamine and TNF-α suppression was calculated from these data.Results: 1.Pae and Oxy of different concentration suppressed the proliferation of RBL-2H3 mast cells in high concentration, and its IC50 were 0.2200mg/ml,0.9454mg/ml,respectively. 2. The percentage of histamine released from RBL-2H3 mast cells activivated by compound 48/80(10μg/ml) was (23.14 + 1.01)% at 30min, which was the pinnacle of the histamine released from RBL-2H3 mast cells. The TNF-α released from RBL-2H3 mast cells activivated by compound 48/80(10μg/ml) was (67. 65±5. 33)pg/ml at 60min, which was the pinnacle of the TNF-α released from RBL-2H3 mast cells. 3. The suppressed percentage of the histamine released from RBL-2H3 mast cells of Pae and Oxy was higher than sodium cromoglicate control group(P<0. 05), The suppressed percentage of the TNF-α released from RBL-2H3 mast cells of Pae and Oxy was higher than sodium cromoglicate control group(P<0. 05) .The suppressed percentage of the histamine and TNF-α released from RBL-2H3 mast cells released was in a concentration dependent manner upon pretreating with Pae and Oxy, The suppressed percentage of the histamine and TNF-α was directly correlated with the concentration of Pae and Oxy.Conclusion: Pae and Oxy may suppress degranulation of RBL-2H3 cell in a direct way, which could be one of the mechanism of antian- aphylactic reaction.
