[Background and Aims] The irritable bowel syndrome(IBS) is a frequent functional bowel disorder in the world. So far the pathogenesis of IBS is still not clear. As we know, it is extremely difficult to obtain the tissues of IBS patients for the study. Therefore, it is significant to set up a animal model of IBS for the study of its pathogenesis. In this experiment, our established rat model of constipation-predominant IBS(C-IBS) was repeated and established by the irritation of cool water intubed to the rat stomach. Using this model, we investigate the changes of interstitial cells of Cajal(ICC), mast cell(MC) and 5-hydroxymetryptamine(5-HT) of the bowels, the aim is to search new clues or evidences for the IBS pathogenesis.[Methods] 56 Wistar rats were divided into 3 groups randomly. The A group, including 24 rats and divided into 3 subgroups A1 , A2 and A3, in each subgroup 8 rats were intubed with icy water for 4 weeks. A1, A2 and A3 group were observed for 4 weeks, 6 weeks and 8 weeks respectively. In the group B, 8 rats were intubed with icy water for 2 weeks and were observed for 6 weeks, In the group C, 24 rats were divided into 3 subgroups in which 8 rats were obversed as control group.The granule number and the moisture content of the feces from the A, B and C3 groups were measured daily. The rectal balloon dilation were performed to evaluate the perception thresholds by the abdominal withdrawal reflex scores of the A and C group. The colon samples from ileocecal junction and distal colon were stained by toluidine blue for mast cells and by immunohistochemistry for 5-HT immunoreactive cells.
