Experiment â… The effects of Propofol on oxidative stress in rats with hepatic injury by the hemorrhagic shock with endotoxinObjective To investigate the effect of Propofol on oxidative stress in rats afterhemorrhagic shock with endotoxin and mechanism.Methods Fifty-six healthy SD rats were used to establish the rat model ofhemorrhagic shock with endotoxin.Male Sprague-Dawley rats were randomlyallocated into following groups (n=8 in each) : pro-traumatic group;sham-operation 2h group; sham-operation 4h group; hemorrhagic shock withendotoxin 2h group(2h traumatic group); hemorrhagic shock with endotoxin 4hgroup(4h traumatic group); Propofol treatment 2h group(PPF 2h group); Propofoltreatment 4h group(PPF 4h group).In the Propofol treatment group,animalsreceived Propofol intravenously at the rate of lOmg/kg/h constantly.The levels ofMDA and SOD in hepatic tissue and the serum concentrations of ALT weremeasured in every rat..Results The mean levels of MDA and ALT elevated after hemorrhagic shockwith endotoxin.The values of 2 hours and 4 hours after giving LPS weresignificantly higher than that of pre-trauma (P<0.05) .Compared with traumaticgroup,there were lower levels of ALT and MDA (P<0.05) and a higher level ofSOD(P<0.05) in the Propofol group and there was attenuated histological changeof liver in the Propofol group.Conclusion Acute hepatic injury after hemorrhagic shock with endotoxin maybe mainly induced by oxidant stress.Propofol may attenuated hepatic injury byhemorrhagic shock with endotoxin due to its antioxidation. Experiment â…¡The effect of Propofol on the expression of Apoptosis Factors in hepatic injury by the hemorrhagic shock with endotoxin in ratsObjective To investigate expression of apoptosis-related proteins in liver after hepatic injury by the hemorrhagic shock with endotoxin,and to measure the modulating effect of Propofol on the experssion .Methods We establish the rat model of hemorrhagic shock with endotoxin in 56 healthy SD rats. Male Sprague-Dawley rats were randomly allocated into following groups (n=8 in each) : pro-traumatic group; sham-operation 2h group; sham-operation 4h group; hemorrhagic shock with endotoxin 2h group(2h traumatic group); hemorrhagic shock with endotoxin 4h group(4h traumatic group); Propofol treatment 2h group(PPF 2h group); Propofol treatment 4h group(PPF 4h group).In the treatment group,animals received Propofol intravenously at the rate of 10mg/kg/h constantly. The expression of Bax and Bcl-2 in heaptic tissue was measured and calculated by immunohistoche analysis in different observation time:pre-trauma, 2 hours and 4 hours after giving LPS. Results (1)Histological dammage occurred after trauma,and it was charactered by hepatocytic swelling or necrosis.These changes were markedly alleviated in treatment group.(2)The expression of Bax and Bcl-2was showed in all animals. At traumatic group the expression of Bcl-2 elevated at the 2 hour group and 4 hour group after giving LPS ,which were significantly higher than that of pre-trauma group (P<0.05) . Compared with traumatic group, higher expression occured in the Propofol group (P<0.05). While the experssion of Bax elevated rapidly,statistic significance was found between the 2h group and 4h group after giving LPS(P<0.05).The experssion of Bax and Bcl-2 keep stable in control group. The values of PPF treatment group were in the middle of the sham-operation group and traumatic group .(3)In traumatic group ,Bax/Bcl-2 showed significant increase at 2h group and 4h group,but there was no changes in other groups. Histological change of liver was attenuated in the Propofol group. Conclusion Following trauma and infection,the expression of Bcl-2 isdecreased,while the expression of Bax is elevated. Propofol may attenuated hepatic injury by hemorrhagic shock with endotoxin through the effect of regulating the expression of apoptosis-related proteins.
