Object: To investigate the effects of valsantan and atorvastatin on the expression of ADMR mRNA and protein in renal injury of diabetic rat, and study the renal protective mechanism of the two medicaments.Methods: Forty weight-matched male Sprague-Dawley rats were randomly divided into following groups: normal rats for control(n=10), STZ-induced diabetic rats(n=10), diabetic rats treated with valsantan (n=10) and diabetic rats treated with atorvastatin(n=10). Diabetic rats were induced by single intraperitoneal injection (70mg/kg body wt) of streptozotocin(STZ). STZ-induced diabetic rats were treated with valsantan (10mg/kg body weight), atorvastatin (15mg/kg body weight) or vehicle for 8 weeks. Then we tested weight, blood sugar, HbA1c and UAER. The expressions of ADMR mRNA in renal cortex were analysed by RT-PCR, as well as ADMR protein were detected through Western blot.Results: (1) Compared with the normal, the body weigh of diabetics was sharply declined ,while blood sugar, HbA1c and urinary albumin excretion(UAER) in 24h were increasing. Atorvastatin and valsantan treatments reduced urinary albumin excretion in 24h, while valsantan treatment increased body weight, compared with the untreated.But no notable difference was seen in HbA1c and blood sugar of diabetic rats between the two groups. (2) Expressions of ADMR in renal cortex of diabetic rats were much more than that of the control. Valsantan and atorvastatin treatments increased the expressions of ADMR mRNA and protein in diabetic rats renal cortex.Conclusions: (1) Valsantan and atorvastatin treatments can alleviate renal injury of diabetic rats, but have no effect on blood sugar and HbA1c. In addition, valsantan can increase the body weight of diabetic rats. (2) Valsantan and atorvastatin treatments can sharply increase the mRNA and protein expressions of ADMR in renal cortex of diabetic rats. (3) Our findings suggest that the beneficial effect of the two medicaments might be mediated by ADMR.
