ObjectiveTo compare the effect of glucose in high or low concentrations on the expression of extracellular matrix in mesangial cells in vitro, investigate the effect of complement factor H and adrenomeduUin on rat mesangial cells cultured under high glucose condition, and observe the effect of diovan, atovastatin and pioglitazone on the expression of complement factor H and adrenomeduUin.MethodsThe mesangial cells cultured in medium in vitro under low (glucose: 5.6mmol/l) or high glucose concentration (glucose: 30mmol/l) were treated by different concentrations of complement factor H, adrenomeduUin, diovan, atovastatin and pioglitazone. ELISA method was used to measure the content of TGF-β1 in the supernatant of cultured mesangial cells, and RIA method was used to assess the content of laminin and type IV collagen. RT-PCR was applied to determine the expression of TGF-β1mRNA, AMmRNA and CfhmRNA. Western blot was performed to measure the expression of adrenomeduUin and complement factor H.Results1. The contents of TGF-β1, LN and type IV collagen in the mesangial cells cultured under high glucose conditions were significantly increased, while adrenomeduUin and complement factor H could significantly decrease their expression in a dose- and time-dependent manner for a certain extent.2. After the intervention of diovan, atorvastatin andpioglitazone, the contents of TGF-β1, LN and type IV collagen in the extracellular matix of mesangial cells cultured under high glucose conditions were significantly reduced compared with high glucose control group. And also, the expression of TGF-β1mRNA, AMmRNA and CfhmRNA were significantly decreased compared with high glucose control group. And in the same time, the expression of AM and Cfh were also reduced.Conclusions1. adrenomedullin and Complement factor H could inhibit the over-expression of extracellular matrix in mesangial cells induced by high glucose, which showed the renal protective functions.2. Diovan, atorvastatin and pioglitazone all showed renoprotective functions in a certain extent, which mechanism might concerned with the inhibition of TGF-β1 expression. After the intervention of diovan, atorvastatin and pioglitazone, the expressions of AMmRNA and CfhmRNA were reduced, which were might also related to the variation of TGF-pi.
