Objective: To provide satisfactory transplant materials and methods for reconstruction of the corneal surface, utilizing the tissue engineer technique to rebuild the limbus corneal epithelial in vitro. Methods: The limbus corneal tissues selected from healthy eight newborn and eight adult white New Zealand rabbits were taken by surgical microscopy, then cultured to reconstruct corneal epithelial layer. The tissue grafts from left eyes of newborn and adult rabbits were seeded onto fibrin glue films mingled with equivalent Thrombin and Fibrinogen, and the other grafts from their right eyes seeded onto common cover glasses. Both newborn and adult rabbits' cell growth roofing area were recorded daily. Also, their cells' characters under light microscope, ultrastructure characters and immunocytechemistry behavior were observed, measured and compared .The limbus corneal epithelial growth difference on fibrin glue between the newborn and the adult rabbits was calculated by paired t test. The adult rabbits' limbus corneal epithelial growth difference cultured on fibrin glue and cover glass slides was compared by the double-independent sample t test, respectively.
