Objective:Simple obesity is a chronic disease, in which energy balance is broken—energy intake excede energy expenditure, the excess energy is stored in the form of fat. It is related to many diseases which impact people's life seriously, such as type 2 diabetes(T2DM), atherosclerosis etc, and has no effective treatments. Adiponectin, a protein secreted especially by white adipose tissue, has been reported to have an important role in regulating energy balance and the sensitivity of insulin. We studied the variety on adiponectin expression levels in obesity and T2DM patients, and discussed the role of adiponectin in the occurrence and development of simple obesity and T2DM.Now, most results on adipose cell were based on the 3T3-L1 cell line which is derived from mouse embryo. This cell line can not represent the human adipose cell exactly and there were no report on establishing of human adipose cell line. We discussed the appropriate program of the primary cell culture and differentiation of human adipose stromal cell. By this way, the adipose cell differentiated from adipose stromal cell will be aideal model for adipose metabolism and endocrine function study, and will give more means for investigating the related diseases as obesity, diabetes etc, in depth. Methods:1. Collect the serum samples of simple obesity patients and T2DM patients, use ELISA to detect the adiponectin contents in those serum samples.2. Primarily culture and differentiate the human adipose stromal cells, evaluate the conversion process of adipose stromal cells by oil red O staining.Results:1. Serum adiponectin levels in simple obesity and T2DM were significantly lower than those in normal control(P<0.01). Adiponectin in T2DM with obesity also significantly lower than that in T2DM without obesity(P<0.01).2. Line correspondence assay showed that BMI, waist circumference, WHR, FBS, FINS, HOMA-IR were all negatively associated with adiponectin level(P<0.01), but no association was observed with TC, TG, HDL and LDL.
