Salivary adenoid cystic carcinoma(ACC) has a characteristic of perineural invasion, so there is great clinical significance to seek the pathological histologic base of perineural invasion in ACC and understand the reasons for perineural invasion in ACC. In the prior research, our research team conjectured that there may be Schwann cell differentiation in onco-myoepithelial cell of ACC, and it may be the pathological histologic base of perineural invasion in ACC. They also conjectured that neurotrophins and their receptors may be biological markers for perineural invasion in ACC and enhanced expression of them may contribute to perineural invasion in ACC. Objective: To further investigate that there may be Schwann cell differentiation in onco-myoepithelial cells of ACC, and it may be the pathological histologic base of perineural invasion in ACC. To further investigate that neurotrophins and their receptors may be biological markers for perineural invasion in ACC and enhanced expression of them may contribute to perineural invasion in ACC. To construct gene expression profilings on perineural invasion in ACC. To provide evidence for the perineural invasion in ACC bystudying on the pathological histologic base of perineural invasion in ACC.Methods: 1. Double-label immunofluorescence and CLSM was used to detect the expression of Leu-7 and GFAP, which are both Schwann's cell markers, and the expression of a-SMA, which is a new kind of myoepithelial cell marker in salivary ACC tissue samples. Leu-7, which is a new kind of Schwann's cell marker, was examined in salivary ACC tissue samples by immunohistochemical method and image analysis. The results were compared with normal salivary tissue samples and acinic cell carcinoma tissue samples which don't have the character of perineural invasion. To analyze the relationship of the expression of Leu-7 and perineural invasion by combining clinical data. 2. TrkB, which is a high-affinity neurotrophin receptor, was examined in salivary ACC tissue samples by immunohistochemical method and was analyzed the relationship of the expression of TrkB and perineural invasion. 3. To procure the pure tumor cells of ACC which have invaded the nerves by using laser capture microdissection(LCM) and RT-PCR technic, and to further construct gene expression profilings on perineural invasion in ACC by using gene expression profiling microarrays.Results: 1. The capability adenoid cystic carcinoma on neural invasion is more higher than acinic cell carcinoma, there were significant differences(P=0.016). Leu 7 expressed in most of adenoid cystic carcinoma, but in none of acinic cell carcinoma, there were si gnificant differences(P=0.004). 2. In salivary ACC tissue samples, the Schwann's cell markers Leu-7 ,GFAP and the myoepithelial cell marker a-SMA proteins were coexpressed in kytoplasm of the same myoepithelial cells. 3.
