ObjectiveQuantitive evaluation of cytotoxicity and sensitivity of paclitaxel against human colorectal cancer cells CX1. And to investigate the effect of MAPK/ERK ki-nase( MEK) inhibitor,PD98059, on apoptosis of CX1 cells induced by Paclitaxel.MethodsCX1 cells was treated by PD98059 and Paclitaxel with different concentration. Methyl thiazolyl tetraolium( MTT) assay was used to determine the inhibitory action . The cells cycle analysis was determined by flow cytometric to study apoptosis of CX1 cells which were exposed to Paclitaxel and PD98059.ResultsPaclitaxel showed strong cytotoxicity on CX1 at IC502. 14 10-7mol/L. A combination of paclitaxel and PD98059 can significantly enhance inhibitory action on CX1 at IC506. 91 l0-9mol/L. The ratio (31.41% ) of apoptosis cells treated with Paclitaxel and PD98059 was significantly higher than that of those (15.6% ) treated with only Paclitaxel (P <0.05).
