Kangshouling (consisting of Radix Polygoni Multiflori Preparata, Radix Astragali, Radix Codonopsis, Radix Angelicae Sinensis, Ganoderma, Rhodiola crenulata, et al) produced good therapeutic effect on Alzheimer' s disease. In order to elucidate the mechanisms of Kangshouling on learning and memory, the effect of Kangshouling on scopolamine-induced amnesia rats and on hydrogen peroxide-induced cultured SH-SY5Y cells are obeserved.In this paper, effect of Kangshouling on scopolamine-induced amnesia in rats is evaluated by Morris water maze test. The results exhibit that 30-days administration with Kangshouling to rats attenuate memory impairment by scopolamine, indicated by shortening escape latency, swimming distance and the time for first reaching the platform(.P<0.05,P<0.01,P<0.01), increasing the swimming time of amnesia rats in the quadrant where platform was placed and frequency to passing through the position where platform was placed(P<0.05,P<0.05). The results also indicate scopolamine, a traditional muscarinic receptor antagonist, evaluated oxidative status in brain characterized by increasing the content of MDA(P<0.01) and inactivation of SOD and GSH-Px(P<0.05,P<0.05) and consequently inhibition of ATPase (P<0.01)in the hippocampus and cerebral cortex in amnesia dose. But Kangshouling reversed it by lowering MDA(P<0.01) and increasing the activities of SOD and GSH-Px(P<0.05,P<0.05) and also enhancing ATPase activity(P<0.01).To investigate protective effect of Kangshouling on the damage of neuroblastoma SH-SY5Y cells cultured in vitro introduced by H2O2, we observed that the medical serm of Kangshouling can attenuate the damage of neuroblastoma SH-SY5Y cells by increasing significantly the survival rate(P<0.01), decreasing the changes in cell morphology, and lowering LDH release from cells into culture medium(P<0.01), suggesting a cognitive improvement of Kangshouling, at least partially, via its antioxidant activity.In conclusion, Kangshouling improve the ability of learning and memory of scopolamine-induced amnesia rats and its related mechanisms at least relevant to its antioxidant effect. In addition, The medical serm of Kangshouling can attenuate the damage of neuroblastoma SH-SY5Y cells which maybe also related to its improvement in learning and memory.
