Morris water maze experiment for a second reference memory of mice, analysis the screening of mice water maze experiment will affect the second water maze experiment, such as in the other direction, have an impact on the platform experiments were made to eliminate the screening for the impact of water maze experiment.Method:ICR mice (male and female half) were randomly divided into training twice a day group and training three times a day, for15consecutive Morris water maze navigation experiments, the experimental repetition of three groups of mice, review the escape latency of mice, target quadrant percentage, length, outer ring indices such as residence time percentage, find out each time point of the specific conditions of the study result in mice, to determine at what point in time in mice remember platform location; Remember the position of the platform in mice, according to the study result in mice were randomly divided into four groups, respectively in15d,30d,45d,60d navigation experimental study on the change law of reference memory in mice; If still remember the position of the platform, in mice within60d reverse experiment platform, investigation and learning ability in mice.Result:Navigation experiment:two times/day is compared between groups of three groups of mice have no significant difference (p>0.05),3times/day is compared between three groups have no significant difference (p>0.05); Three groups of mice swimming speed is no significant difference between groups (p>0.05); Outer ring retention time percentage gradually decrease in mice; Target quadrant residence time percentage, and three times/twice/group d d comparison between the way there is no significant difference (p>0.05); Escape latency, and three times/twice/group d d comparison between the way there is no significant difference (p>0.05); Escape the incubation period is less than70s two percentage comparison, twice/group d after training at the end of the ninth, escape the incubation period is less than70s two percentage reaches80.25%. Three times/d group after training at the end of the ninth, escape the incubation period is less than70s two percentage reaches80.28%. The remaining20%of the mice after9~15times training will remember platform position gradually, and a single appears twice in less than70mice s percentage below5%; Reference memory investigation:in15d,30d,45d,60d escape latency in mice with the test results basic close before, no significant difference (P>0.05); Reverse platform experiment:outer ring length percentage comparison, navigation experiment and reverse platform was no significant difference (P>0.05), reverse platform experiments in mice is a the significant higher percentage of target quadrant residence time navigation target quadrant, length percentage of experiments in mice (P<0.05). reverse platform experiments in mice escape latency significantly below the navigation escape latency in mice (P<0.05), escape the incubation period is less than70s, two in the navigation experiments after the seventh experiment in mice, mice incubation period of two consecutive below70s percentage of80%or more, in reverse platform experiment, after the fifth experiment in mice, mice incubation period of the percentage of two consecutive below70s reached more than80%.Conclusion:Training twice a day and a day training three grades of the mice did not differ two training methods have no difference; More than80%of the mice after nine training can remember where platform:The reference memory in mice within60days no change; In reverse platform experiments in mice can quickly remember the position of the platform.
