Objective To study the role of p21WAF1 and p53 protein in the occurrence and development of large intestine cancer. Methods p21WAF1 and p53 protein were examined by immunohistochemistry SP in 41 example specimen of large intestine carcinoma and 10 normal specimen. DNA content of corresponding specimen were examined by flow cytometry. Results 1.The SPF gradually increase among normal mucosa, well differentiated adenocarcinoma, moderately differentiated adenocarcinoma, poorly differentiated adenocarcinoma , and the difference is significant .2. The SPF in Dukes A+ B phase of large intestine adenocarcinoma is much lower than those in Dukes C+D phase . 3. The expression rate of p53(+) in large intestine adenocarcinoma is 68.2%, and the expression rate of p21 WAF1 (+)is 36.6%. In addition ,there is negative correlation between the expression of p21 WAF1 and the expression of p53. 4. The SPF in large intestine adenocarcinoma - which express both P53(-) and p21 WAF1 (+) is much lower than those in large intestine adenocarcinoma which express both p53(+) and p21WAF1(-). 5. The expession of p21 WAF1 gradually decrease among normal mucosa .well differentiated adenocarcinoma, moderately differentiated adenocarcinoma,poorly differentiated adenocarcinoma .In the meanwhile ,there is correlation between the expession of p21 WAF1 and cell differentiation . 6. The expression of p21 WAF1 in large intestine adenocarcinoma ( Dukes A+B phase) is much higher than that in large intestine adenocarcinoma (Dukes C+D phase) . 7. The expession of p53 gradually increase among normal mucosa,well differentiated denocarcinoma, moderately differentiated adenocarcinoma,poorly differentiated adenocarcinoma . At the same time ,there is correlation between the expession of p53 and cell differentiation . 8. The expression of p53 in large intestine adenocarcinoma of Dukes A+B phase is much lower than that in large intestine adenocarcinoma of Dukes C+D phase. 9. SPF has no significant difference respectively in patients with different age,sex and the location of tumor. 10. The expression of p21 WAF1 and p53 has no significant difference respectively in patients with different age,sex and the location of tumor. Conclusion p21 WAF1 and p53 played an important role in tumorgenesis and development of large intestine adenocarcinoma . p21 WAF1 ,p53 and DNA content could be used as indicators in evaluating prognosis of large intestine adenocarcinoma.
