Objective To investigate the different expression and clinical significance of mi R-21 and mi R-146 a in tissues and serum samples of colorectal neoplasms.Methods The endoscopic biopsy tissues and serum samples of 100 colorectal cancer(CRC) patients,80 colorectal adenoma(CRA) patients and65 healthy controls were collected. The expressions of mi R-21 and mi R-146 a were detected by quantitative reverse transcription polymerase chain reaction. The relationship between the expression of mi RNAs and clinicopathological features were analyzed. Then,the diagnostic values of serum-specific mi RNAs for colorectal neoplasms were evaluated.Results The expression of mi R-21 were confirmed to be significant higher in tissues and serum samples of CRC and CRA patients than in healthy controls(P<0.001). The expression of mi R-21 in CRCs was notably higher than in CRAs(P<0.05). High mi R-21 expression in CRC patients was strongly associated with advanced TNM stage and lymph node metastasis. The expression level of mi R-21 in CRA patients was correlated to histological types(P<0.05). There was a statistically positive correlation between mi R-21 expression in CRC tissues and matched serum samples.Compared with healthy control subjects, the expression of mi R-146 a in CRC patients were decreased(P<0.01). No statistically significant differences were observed in mi R-146 a expression between CRA patients and healthy controls(P>0.05). The expression of mi R-146 a in CRC was reduced compared with CRAs(P<0.01). Furthermore,the reduced expression of mi R-146 a in CRC patients was correlated with differentiation and TNM staging. Nevertheless, no significant correlation was found between mi R-146 a and clinicopathological features including age,gender,adenoma location,adenoma size,histology and CEA in CRA patients(P>0.05). Serum mi R-21,mi R-146 a and combined levels robustly distinguished(AUC=0.889,0.791,0.863) CRC and(AUC=0.784,0.692,0.761)CRA patients from controls. And they yielded AUC of 0.705,0.820 and0.713 in discriminating CRC patients from CRA patients.Conclusions mi R-21 and mi R-146 a have potential values for early detection of colorectal cancer.
