Objective To study the possibility of reversion and mechanisms of arsenic trioxide (As2O3) on human gastric carcinoma cell line SGC7901 in multi-drug resistance. Methods The resistant to ADM of SGC7901/ADM and non-ADM resistant human gastric carcinoma cell line SGC7901/S was used in this vitro study, Expression of P-giycoprotein(P-gp) in ceils was detected by Immunocytechemical assay and Western Blot when arsenic trioxide was used or not, And the effect of the expression of arsenic trioxide on ERK1/2 and Ras was detected by immunocytechemical assay. Result The expression of P-gp was significantly higher in SGC7901/ADM than in SGC7901/S(lmmunocytechemical assay: t=11. 139, P<0. 001; Western blot:t=20. 556, P<0. 001) ; Arsenic trioxide could decrease the expression of P-gp, The dosage was higher and the time was longer and the power of the reversion was better (P<0. 01); Arsenic trioxide could significantly decrease the expressions of Ras and ERK1/2 of SGC7901/ADM (P<0. 01). Conclusion 1. One of the reason of the resistant to ADM of SGC7901/ADM was the higher expression of P — gp; 2. Arsenic trioxide could decrease the expression of P-gp, The dosage was higher and the time was longer and the power of the reversion was better;3. Arsenic trioxide may be related with the signal passageway of Ras—MAPK.
