Objective To research the induction of tumor necrosis factor-alpha (TNF- α ) to retina ganglion cells (RGCs) and the expression of apoptosis-related genes(bcl-2,Bax) in this course. Methods 30 healthy adult Wistar rats were chosen and randomly divided into 5 experimental groups and 1 control group with 5 rats every group. The one random vitreous cavity of every animal of experimental groups was individually injected into 2 μ 1 TNF- α of concentration of 2.5 μ g/ml, 5 u g/ml, 10 μ g/ml, 20 μ g/ml and 40 μ g/ml. The one random vitreous cavity of every rat of control group was injected into 2μ 1 normal saline. They were all sacrificed 2 weeks later, and every eye was enucleated and the retinal tissues were prepared into one set of paraffin sections and one set of electron microscope specimens using routine procedures. The paraffin sections for examination under light microscope were made for HE stain and at the same time they were stained by immunohistochemical method and analyzed positive response intensity in ganglion cell layer by Vidas image analysis system. The data were analyzed using variance analysis and t test by SPSS12.0 statistical software. Results 1.
