Objective To study the effect of Okam J201 on bleomycin induced pulmonary fibrosis in rats and its significance.Methods Pulmonary fibrosis was induced in Wistar rats by intratratrachealinstillation of bleomycin. Then the rats received daily Okam J201 25mg/kg(groupl), 50mg/kg(group2),or normal saline(group control )orally. Normal controls received normal saline both intratracheally and orally. Eight rats in each group were sacrificed 28 days after intratracheal instillation. Histological changes of the lungs were evaluated by HE stain, the expression of Collagen-Ⅰ, Collagen-Ⅲ, tumor necrosis factor (TNF-α) , transforming growth factor β (TGF- β 1 )and platelet derived growth factor-B (PDGF-B) , vascular endothelial growth factor (VEGF) proteins was assessed by immunohistochemistry.Results Pulmonary fibrosis of group2(50mg/kg) was improved as comparedwith that of group model . Protein expression of Collagen-Ⅰ, Collagen-Ⅲ, TNF-α, TGF-β1 and PDGF-B > VEGF in group2 was lower than that in group model. Protein expression of Collagen-Ⅲ, PDGF-B , VEGF in group 1 ( 25mg/kg) was lower than that in group model, protein expression of Collagen-Ⅰ, TNF-α, TGF-β1 in group1 has no difference with that in group model.Conclusion Okam J201 alleviates bleomycin induced pulmonary fibrosis inrats. Inhibiting the expressions of Collagen-Ⅰ, Collagen-Ⅲ, TNF-α, TGF- β1 and PDGF-B , VEGF in lung tissues may be one of the mechanisms.
