Objective: To study the effects of interleukin2 (IL-2) on proliferation and collagen synthesis by rabbit lens epithelial cells ( RLECs) in vitro,and their role in posterior capsular opacification(PCO). Methods: 1.RLECs were cultured in vitro and the morphological changes were observed with light microscope. 2.The effects of IL-2 at different concentrations on proliferation of RLECs in vitro was measured with MTT staining colorimetry and cell count technique; The effects of IL-2 at different concentrations on collagen synthesis of RLECs in vitro was detected with hydroxy proline(HP) colorimetric analysis.Results: 1. The result of cell count technique showed that IL-2 at the concentration of 10U/ml can't promoted RLECs proliferation at the time of 24h, 48h, 72h. (p>0.05) ; IL-2 at the concentration of 100~10000U/ml can significantly promote the RLECs proliferation at all of the three times (p < 0. 05) ;The effect of proliferation was most significant at the time of 48h,while at 72h,the effect began to decrease.At the same time,the effects of proliferation on RLECs of IL-2 at all of the experimental concentrations is significantly(p < 0.01),it showed that the proliferation of IL-2 on the RLECs was dependented on the concentrations.2. The result of MTT was consistent to that of cell count technique.3. IL-2 can increase collagen synthesis at all of the experimmmental concentrations (p < 0. 05). The effect of increasing collagen synthesis was significantly,and was depended on the concentrations.Conclution: IL-2 can promote RLECs proliferation and collagen synthesis and can contribute to the PCO by this way. It may be one of the methods to take measure to inhibite the effects of IL-2 during or after the operation of cataract to prevent PCO.
