Objective: to study the effects of Gemcitabine on prohibition by rabbit lens epithelial cells (RLECs) in vitro, and to investigate their role in posterior capsular opacification (PCO).Methods: RLECs were cultured in vitro and the morphological changes were observed with light microscope.The effects of Gemcitabine at different concentrations on prohibition of RLECs in vitro was measured with MTT staining colorimetry and cell count technique.Results: 1. The result of cell count technique showed that Gemcitabine at the concentration of 64 μg/ml can't prohibit RLECs at the time of 24h(P>0.05);Gemcitabine at the concentrations of 128 μ g/ml~4096 μg/ml can significantly prohibit the RLECs at 24h,48h,72h (P<0.05).The effects of prohibition of Gemcitatabin at the concentrations of 1024 μg/ml,2048 μg/ml, 4096μg/ml can significantly prohibit the RLECs compared with the concentrations of 128μg/ml,256μg/ml,512μ g/ml at all of the three times,it showed that the prohibition of Gemcitabine on the RLECs was depended on the concentrationsAt the same concentrations the effects of prohibition on RLECs were depended on times.2. The result of MTT was consistent to that of cell count technique.Conclusions: Gemcitabine can prohibit the RLECs,and can prohibit posterior capsular opacification.Because the circumstances between RLECs and HLECs are different, we need more informations about Gemcitabine on HLECs.Gemcitabine may be one of the medicine to prevent posterior capsular opacification.
