Objective: To establish hyperuricaemia models in rat and to observe the hypouricemic effect of hypouricaemic mistura on the serum uric acid level in hyperuricaemia rat.Methods: Forty-eight male Wistar rats (180~220g ) were randomized to six subgroups: the controls(group A ), hyperuricaemia not be treated group (group B), hypouricemic mistura (low doses, medium doses and high doses) groups(group C, D, E), benzbromarone group(group F). All the drugs were given orally for four days and the animal model of hyperuricaemia were established on the fifth-day of giving drugs:the rats in group A were treated with sodium carboxymethyl cellulose and the rest with the uricase inhibitor potassium oxonate by intraperitoneal injection to increase the serum urate level. The whole blood samples were collected from rats by tail vein bleeding one hour after final administration.. The serum uric acid was determined by uricase-EHSPT. The difference of the data was analyzed by analysis of variance (ANOVA).Results: The serum uric acid level of the hyperuricemic groups were all significantly higher than the controls (P < 0.01) . The serum uric acid level of groups C,D,E and F.which had deserved treatment ,were lower than that of group B, which had not been treated, but only the groups E and F were significant (P < 0.05, P < 0.01) . The serum uric acid level of group F was lower than those of groups C,D and E,but was not significant (P>0.05) .Conclusion: The uricase inhibitor potassium oxonate can increase the serum urate level significantly.Hypouricemic mistura possesses in vivo potent hypouricemic effect on the serum urate level and was dose-dependent.
