OBJECTIVE To examine the different expression of nuclear transcript factor Kappa B (NF-KB) in human lens epithelial cells between the group with Age-related Cataract and the control group. To examine the different expression of NF-KB in human lens epithelial cells between the group with Traumatic Cataract and the control group. And to examine the different expression of NF-KB in human lens epithelial cells between the group with Congenital Cataract and the control embryo group.METHODS The anterior capsules of lens with Age-related Cataract, that with Traumatic Cataract, that with Congenital Cataract and that of normal corpse eye donor were collected by microscope and preserved in -180 °C fluid nitrogen. Total RNA was extracted from anterior capsular lens tissues, and as a template, cDNA was synthesized by Oligo(dT) primer. Specific nuclear factor Kappa B p65 primers were derived from Gene Bank (L19067) to amplify NF-KB p65 and house keeping gene β-actin specified DNA fragment by polymerase chain reaction (PCR). According to kinetics experiments, we chose 26 cycles for β -actin and 29 cycles for NF-KB. PCR products were performed by 2%-3% agarose gel electrophoresis and were analyzed by BiocapMw in order to acquire relative quantity of NF-KB PCR products. Using SPSS (version 11.5) Independent-Samples T test to analyze the statistic. Significance was considered as P<0. 05.RESULTS Compared with the mean expression on normal control group , the mean expression of nuclear factor Kappa B in the anterior capsular lens epithelia of the group with Age-related Cataract is 0.9998, which is higher than that in control group 0. 8337, and significance is P<0. 05 based on Independent-Samples T test (T=4. 016) .Compared with the mean expression on normal control group,...
