Objective: To evaluate the association between cytokeration 19(CK19) mRNA and cytokeration 20(CK20) mRNA with the micrometastasis in the circulating cells of breast cancer by fluorescent quantification polymerase chain reaction (FQ-PCR) .Methods: CK19 and CK20 mRNA in the of clinical samples of 15 health women ,20 patients with benign breast disease and 31 patients with breast cancer were detected by FQ-PCR. GAPDH was used as internal controls.The measurement data was analyzed statisticly by Q-test and enumeration data by chi-squere test.Results: There were no significant difference in CK19 mRNA and CK19/GAPDH between normal controls and benign breast disease group(p>0.05). CK19 mRNA and CK19/GAPDH in breast cancer group were higher than those in the other groups (p<0.05). There were no difference in GAPDH among three groups (p>0.05). The percent of positive of CK19 mRNA and CK19/GAPDH in 31 breast cancer patients were 61.3%(19/31) .In the detection of cytokeration 20(CK20) mRNA ,there were no significant difference among three groups (p>0.05), and the percent of positive of CK20 mRNA and CK20/GAPDH in 31 breast cancer patients were 3%(1/31 ). There were no significant difference between pre-operation and 1 week after operation in CK19 mRNA and COO mRNA (p>0.05).The later was lower than the former.Conclusions: FQ-PCR is a rapid ,sensitive and specific method for quantitating CK19 and CK20 mRNA .The detective of CK19 mRNA gives objective evidence to the micrometastasis of breast cancer .
