Objective: to investigate the changes of serum adiponectin and TNFalevels and their relationship with fatty liver in impaired glucose tolerant (IGT). Methods: Eighty male Wistar rats weighing about 180g were dividedrandomly into two groups. There are twenty in normal control (NC) and forty inmodel group (MG). The MG were fed with high-fat feeding.8weeks, 12weeks,20weeks after high-fat feeding, the blood samples were obtained to measure thelevels of adiponectin ,TNFa ,TG ,FINS, et al. The liver sample were used toanalyse morphologically and homogenized to measured the levels of TG. Wecompared these data between the two groups and among the different stages andreveal the role of adiponectin and TNFa in the pathogenesis of fatty liver in IGT. Result: 1.Compared with the NC,the weight, oral glucose test 2 hour plasmaglucose (OGTT2HPG) were higher in IGT group. There was a significantdifference between two groups (P<0.05-P<0.01). After 12-week high-fat feeding,OGTT2HPG reached 8.58±0.82, the rat model of IGT were produced. Theplasma TG concentrations of rats with high-fat feeding was evidently higher at20 week. 2.The plasma FINS concentrations of IGT group increasingly elevated.Compared with the NC, the difference is significant in 8week,12week,20week. 3.Liver sample were HE-stained. There was evidently fatty infiltration inliver of IGT group. It markedly differed from the NC. Hepatic TG contentsincreasingly elevated in liver of IGT group. 4.the plasma adiponectin concentrations of IGT group increasinglydecreased. In the meantime IAI increasingly decreased too. It indicatesdecreased adiponectin may play an important role in fatty infiltration of IGT. 5.The plasma TNFa concentrations of MG increasingly elevated. The fattyinfiltration in MC was more serious. It indicates increased TNFa may play animportant role in fatty infiltration of IGT. 6.Multiple stepwise regression analysis showed that FIN,TG,adiponectinwere independent risk factor of fatty liver. The decreased adiponectin was themost important factor in fatty infiltration of IGT. Inclusion: 1. we successfully produced IGT model by 12-week high-fat feeding. 2.Decrease adiponectin and increase TNFa correlated closely with thedevelopment of fatty liver, as judged by the TG levels of liver tissuehomogenate.
