Objective: To study the effect of dendritic cells(DC) co-cultured with cytokine induced killer cells(CIK) on cytotoxicity against tumor cells.Methods: Peripheral blood mononuclear cells(PBMC) can be isolated from heathy adult donors. After 2 hours incubation of PBMC, DC can be induced from adherent cells by some cytokines and CIK can be generated from non-adherent cells. Mature DC can be harvested after 9 days incubation, and then co-cultured with CIK at a ration of 1:5. The cytotoxicity activity was investigated in MTT assays against SMMC-7721 hepatocellular carcinoma cell line ,K562 and K562 drug-resistant cell line.Results: CIK cells were able to lyse SMMC-7721 hepatocellular carcinoma cells and K562,K562 drug resistant cell line at low effector to target ratios. This effect was significantly enhanced by co-culturing with DCs. Conclusions: CIK cells showed high lytic activity against SMMC-7721 hepatocellular carcinoma cell line and K562,K562 drug resistant cell line.,which could be enhanced by co-culturing with DC. DC-CIK cells are highly effective immune cells.
