Objective: Uterine leiomyomas are very common benign tumors of procreative organ inwomen of reproductive age.It is estimated that more than 20% of women above the age of 35 have a leiomyoma in china , according to corpse anatomical information.Because many sufferers have no symptoms,or the leiomyoma is very little,clinical incidence reported is far less than that actually.About 50% of the diseasers have symptoms,includeing pelvic cavity pain ,pressure feeling and over uterine bleeding,which seriously affect women"s body health and life quality.There are no satisfactory nonsurgical treatments because of unclear pathogensis of leiomyoma .At present,hysterectomies are the most frequent treatments in the world , and leiomyoma is a major justification.Uterine leiomyoma is considered an estrogen-associated tumor.Evidence supporting this includes its development during the term of procreation ages,enlargement and growth during pregnancy,and regression after menopause.The estrogen functions physiologically by its relative receptors.There are more estrogen receptors and estradiol in leiomyoma than myometrium.Recent research indicates some other hormones and growth factor,such as PR,VEGF and IGF1Rα,may play an important role in the pathogenesis of uterine leiomyoma.To explore the role of ER,PR,VEGF and IGF1Rαin the mechanicsms of the uterine leimoyoma development and to offer more scientific theoretic basis for nonsurgical treatment of uterine leiomyoma. Methods: 1 Sample"s source: Thirty patients with leiomyoma undergoing hysterectomies at the He Bei Province Ren Min Hospital were selected randomly for study subjects in May-October 2003.They receivedno hormonal treatment and have no disease of internal secretion such as diabetes mellitus,hyperthyroidism,hypothyroidism,ovarian tumors and so on during the minimum 6 months period precedeng surgery.Leiomyoma and myometrium specimens were obtained from these patients during hysterectomyies. Leiomyoma tissue samples were collected to be objective group,whereas myometrium tissue samples were collected to be control group. 2 Sample"s collection and disposal: 2.1 Each of fresh tissue samples(1.0cmX1.0cmX0.2cm) including tumor and normal were removed from the center of Leiomyoma or apart from myometrium after uteri were resected from bodies,and were rapidly fixed in 4% paraformaldehyde(ph:7.2-7.4) for 24-48 hours,then dehydrate through increasing concentrations of alcohol(80%-90%-100%),clarified within dimethybenzene,and paraffin wax embedded ,5um serial section,stained byhematoxylin-eosin (HE) and immunohistochemistry(IHC). 2.2 Limosis blood was taken out from leiomyomas sufferers,blood plasma was parted out,and preserved in refrigerator at -20 degree for use. 3 Detect the expression and distribution of ER,PR,VEGF and IGF1Ra on leiomyomas and myometrium in each group through immunohistochemistry SP method. estradiol and progestin was examined through enzyme-immunity method. Results: 1 The result of the IHC analysis reveals that ER was expressed in 28 cases of leiomyomas and 22 cases of myometrium ,among which there were 7 cases of equivocally positive expression ,8 cases of positive expression and 13 cases of extra positive expression in leiomyomas,and there are 10 cases of equivocally positive expression, 4 cases of positive expression and 8 cases of extra positive expression in myometrium.The expression of ER in leiomyoma was higher than that in myometrium(u=2.2557,0.010.25) and myometrium(u=1.1269,0.10
