Objective:Intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1) are members of immunoglobin superfamily adhesion molecules, expressed on the surface of activated endothelial cells, T cells and B cells. ICAM-1 and VCAM-1 participate in many physiologic and pathologic processes, such as inflammation, immunological reaction, immunological responds, tumor metastasis, thrombosis, et al. Recent researches have indicated that ICAM-1 and VCAM-1 are important in the adhesion and interaction between fetus and maternal uterine. Trophoblast cells are a special kind of epithelial cells during implantation and placentation. Their activity to invade the maternal spiral arteries resembles the invasion of tumor cells. The difference is that the invasion of normal trophoblast cells is distinctively limited by time and space. Hypertensive disorders complicating pregnancy is a multisystem disorders of human pregnancy, representing the leading cause of both fetal and maternal morbidity and mortality in pregnancy. Despite intensive efforts to find mechanisms and markers inducing hypertensive disorders complicating pregnancy, no specific etiological factors have been identified until now. Many hypotheses have been described, but none can be confirmed completely. Since Young put forward uterine-placenta ischemia theory in 1918, it has been universally accepted. According to many scholars, hypertensive disorders complicating pregnancy is a placental disease. The first stage is the process that affects the spiral arteries and results in a deficient blood supply to the placenta. The second stage encompasses the effects of the ensuring placental ischemia on both the fetus and the mother. Gestational trophoblastic disease is a group of tumor induced by over-proliferation and excessively invasion of trophoblast cells. With the progress of the disease, trophoblast cells represent the unlimitedly proliferation and invasive character like tumor cells. To investigate the importance of ICAM-1 and VCAM-1 to trophoblast cells, we compared the expression of ICAM-1 and VCAM-1 on trophoblast cells of the first trimester pregnancy villi that is normal or naturally aborted and of hydatidiform mole. We also compared the expression of ICAM-1 and VCAM-1 on trophoblast cells of placenta from normal and preeclampsia third trimester pregnancy. Methods:This study involved two groups. One group consisted of 56 women at the first trimester of pregnancy,20 of them were healthy pregnant women requiring induced abortion,18 complained with unexplained abortion and 18 diagnosed hydatidiform mole by B ultrasound. The villi and hydatidiform mole were taken from them. The other groupconsisted of 20 normal third trimester pregnancy and 18 preeclampsia women. Placentas were taken when they took on caesarean surgery before uterine contraction. All tissues were routinely fixed in 10 percent formalin and embedded in paraffin. Five-μm-thick tissue sections of all samples were stained by immunohistochemical analysis. IOD was used to assess the immunostaining intensity of trophoblast cells. T-test and analysis of variance were used to analyze with SPSS12.0 software, α<0.05 was considered statistically significant. Results:1 The expression of ICAM-1 and VCAM-1 on trophoblast cells of villi and hydatidiform mole was showed by Fig1,3-7 and Table1. The expression of ICAM-1 and VCAM-1 on trophoblast cells of normal villi was the strongest (3.836±0.219,3.962±0.363), and that of natural abortion is almost negative (0.286±0.080,0.230±0.058), hydatidiform mole had weaker than normal villi and stronger than natural abortion expression of ICAM-1 and VCAM-1 on trophoblast cells(1.862±0.162,1.753±0.135).The differences among the three groups were significant (p<0.01). 2 The expression of ICAM-1 and VCAM-1 on trophoblast cells of preeclampsia is weaker than normal placenta (0.287±0.074,1.773±0.125 vs 2.813±0.130,2.750±0.182), The difference was significant (p<0.01). Conclusion:Based on the data mentioned above, we can come to the following conclusions: 1 Normal trophoblast cells can express ICAM-1 and VCAM-1 in early pregnancy. The...
