Defaecated And Poison-Discharged Functional Research Of Active Components From Semen Cassiae

To study the active components from Semen Cassiae and their defaecated and poison-discharged effect, the experiments in this paper were divided into two main parts: the component analysis experiments and the animal experiments. The component analysis experiments were composed of three subparts, that is, the content determination of polysaccharides and anthraquinones from SC â…  and SC â…¡, the HPLC analysis of SC â…  and the SDS-PAGE analysis of SC â…¡. The animal experiments of purgative and poison-discharged effect were divided into four subparts, that is, the purgative experiments of Semen Cassiae;the purgative experiments of SC â…  and SC â…¡;the purgative and poison-discharged experiments of aether extracts and residual of SC â… ;and the microorganism analysis experiments.The results of the component analysis experiments showed that the content of the anthraquinones from SC â…  (the shell of Semen Cassiae) was 1.2027% (from SC â…¡ was 0.4435%), the HPLC analysis of SC â…  indicated that there were emodin, chrysophanol and physcion etc. in the aether extracts, but they could not be made single extracted by aether and acetone alternatively. The content of cellulose from SC â…  was 7.59%. The content of the water-soluble polysaccharides from SC â…¡ (the kernel of Semen Cassiae) was 3.9675% (from SC â…  was 0.3236%). And the SDS-PAGE analysis of SC â…¡ showed that the polysaccharides were neutral and macromolecular.The purgative and poison-discharged animal experiments showed the wet and dry weight, water content and kilo-granule weight of dejections of rats in taken Semen Cassiae were obviously increased (p<0.01) compared with the control group. While the body weight, the intake and the weight of liver and kidney of rats of different groups had no difference. The best add of SC â…  and SC â…¡ in feeds of rats were both 2%, and the two both could increase obviously the granules ,the weight of dejections of rats, and promoted the defecate frequency and improved the properties of dejections. The aether extracts of SC â…  had better purgative effect, but waved greatly and had short period. The purgative effect of residues was steady. And the poison-discharged periods of extracts and residues were about two weeks. The content of ammonia, hydroxybenzene and indole in the dejecta of rats eaten the extracts were the highest. The microbiological experiments told that SC â…¡ could promote the multiplication of probiotic bacteria;While SC â…  did not have such effect.
