Background and Objective:Cerebral infarction is one of the common cerebrovascular diseases. Its physiopathologic basis is the cerebral arteriosclerosis. The "damage reaction theory" is taken as the mail pathogenesis of atherosclerosis. That is, the damage of vascular endothelial cells is the initiation factor of atherosclerosis . Moreover, the adhesion molecule ICAM-1 induce the blood cells accumulation in vascular endothelium and play an important role in cerebral infarction. Many studies indicate that only a little of ICAM-1 is expressed on vascular endothelial cells and other cells under normal conditions. But its expression rises clearly as the tissues are suffered in ischemic and this leads the tissues and cells to damage.hyperuricemia is not only the reason of gout and gouty nephropathy but also one of the important danger factors of atherosclerosis. Urate calculus could damage thevascular endothelium cells when hyperuricemia and promote the atherosclerosis through the purinemetabolism and lipometabolism. Some studies indicate that the level of blood urate is close related to the incidence rate and death rate of acute cerebrovascular diseases(ACVD).The linear relation between hyperuricemia and ACVD is: the higher the blood urate, the heavier the disease and the worse prognosis.Because of the common improvement of people's living standards, diet changes. The intake of food rich in purine and protein is visibly increasing. The incidence rate of hyperuricemia increases with years. So the treating of hyperuricemia is the key aspect of preventing cerebrovascular diseases.Acute cerebral infarction belong to "apoplectic" category in Chinese medicine.The causes include liver wind, fire, phlegmdampness, stagnant blood, qi loss and yin loss et al. Niao Suan Li Xian is composed of greenbrier, large leaf gentian root, milk vetch, root of red-rooted salvia, loranthus, edible tulip and coix seed. It could strengthen spleen and tonify kidney, promot blood circulation to dissipate blood stasis and clear away heat. In clinic, it is proved to have good cure to kinds of ACVD.In our study, we will culture human vascular endothelial cells(EC) induced by urate and examine the expression of ICAM-1 on the surface of vascular endothelial cells to investigate the effect of Niao Suan Li Xian (serum of including drugs) and evaluate the role of ICAM-1 in the pathegenesis of cerebral infarction.Meterial and Methods:1. The human vascular endothelial cells(EC) were quickly diffluenced which were kept in -80℃. The cells' suspension liquid was shift into culture bottles, then put the cells into the 37 ℃, 5%CO2 and culture in way of passage.2. Make the serum of including drugs.ultrafiltrate, load and put in -20°C. Diffluence it in 37℃ when use it.3. The cells were stimulated by urate of three levels(710umol/L, 1070umol/L, 1430umol/L) for 24 hours. The cells of normal group were cultured without urate.4. The cells were stimulated by urate of middle level(about 1070umol/L) for 24 hours and treated with the serum of including drugs. The cells of normal group were cultured only with culture liquid, the model group with urate talked above on basis of the normal group and the treat group with serum of including drugs on basis of model group.5. Draw total RNA out of the cells with TRIZOL. The expression of ICAM-1 mRNA was examined by RT-PCR. The glues were scanned and canculate the opposive quantity of ICAM-1 mRNA. Compare the difference among the groups.Results:1. Compared with the normal group, the expression of ICAM-1 mRNA increased clearly in the model group of low and middle levels urate (p<0.05) and the middle level was better. But in the high level group, the expression of ICAM-1 mRNA decreased.2. Compared with the model groups, after treated with Niao Suan Li Xian (serum of including drugs), the expression of ICAM-1 mRNA decreased (p<0.05).Conclusion:1. The urate could increase the expression of ICAM-1 mRNA, so hyperuricemia may lead to atherosclerosis by way of stimulating the expression of ICAM-1.2. Niao Suan Li Xian (serum of including...
