Background: Endometriosis is a gynecological disorder characterized by the presence of endometriuml glands and stroma outside their normal location within the endometriuml cavity. The etiology of endometriosis is still unknown. Most of studies focus on the difference between the eutopic endometrium and ectopic endometrium of the same patient. Some data show that endometriosis has the characters of familial incline and heredity. It indicates that the differential gene expression between the patients with endometriosis and without endometriosis is one of the most important factors about the pathogenesis of endometriosis. So this paper emphasize on screening out the differential expression genes about eutopic endometrium and identifying them by several kinds of technique, So that we can find out the basic pathogenesis about endometriosis.Part â…  The differential gene expression profilein endometriosis screening out by cDNA microarraysObjective: To investigate differentially regulated genes in endometrium from women with vs. without endometriosis and discuss the pathogenesis of endometriosis. Methods: Analysis of human gene expression of endometrium with endometriosis and without endometriosis in different period of menstrual cycle with 14112 human genes of cDNA microarrays. Results: 193 genes expressed in endometrium were screened out in the period of proliferation. Among them, 58 were up-regulated, 135 were down-regulated; 93 genes expressed in endometrium were screened out in the period of secretion. Among them, 6 were up-regulated, 87 were down-regulated. 20 genes were screened out both in the period of proliferation and secretion. Among them, only 1 gene wasup-regulated, 19 genes were down-regulated. Conclusion: cDNA microarray technique is effective in screening out the differentially expressed genes between normal and abnormal tissues. Part â…¡ Identification of differentially expressed genesabout endometriosisObjective: To identify the differentially expressed genes, tissue inhibitor of metalloproteinase-3(TIMP-3), matrix metalloproteinase-2, 9(MMP-2, MMP-9) and investigate expression of the three in eutopic endometrium from women with and without endometriosis throughout the menstrual cycle. Methods: 1. By immunohistochemical staining, the expression of protein of MMP-2, MMP-9 and TIMP-3 were detected in 60 eutopic endometrium with endometriosis and 72 normal endometrium. 2. By reversed transcription polymerase chain reaction (RT-PCR), the expression of mRNA of MMP-2, MMP-9 and TIMP-3 were detected in 39 eutopic endometrium with endometriosis and 31 normal endometrium. 3. By gelatinase zymography, the activity of protein of MMP-2, MMP-9 and TIMP-3 were detected. Results: 1.We found low positive rate of MMP-2 and MMP-9 (57%, 40% respectively) expression in normal endometrium, while there was high positive rate of MMP-2 and MMP-9 in eutopic endometrium with endometriosis. The proteins of TIMP-3 expressed higher in normal endometrium than eutopic endometrium with endometriosis. 2. Eutopic endometrium from women with endometriosis expressed higher levels of MMP-2 and MMP-9 and lower levels of TIMP-3 than did endometrium from normal women. Endometrial expression of MMP-2 and MMP-9 is elevated during the proliferation phase to secretion phase in normally cycling women while there has no significant difference in the women with endometriosis. 3.
