The purpose of the experiment is to compare the inhibiting scar formation effect of non-penetrating trabecular surgery with amniotic membrane and SK GEL implant in rabbit eyes. We used 18 New Zealand white rabbits and divides them into two groups (12 in group A and 6 in group B). The eyes of these rabbits in each group are randomly defined as experimental group and control group. Amniotic membrane is planted into the eyes of A experimental group and SK GEL into the eyes of B experimental group, nothing implant in control group. The forms of the filter blebs and intraocular pressure (IOP) are investigated every day. One or two typical animals are killed at 1,3,8,12 weeks after the surgery, respectively. Immediately after death the eyeballs are got, 6*6mm surgery area is cut. After fixation in 10% formaldehyde, the samples are dyed by hematowylin and eosin stain, and examined by light microscopy (LM). In addition, one typical animal is killed in group A and B at 3,8 weeks respectively, the samples are fixed in 2. 5% glutaraldehyde for examinationby transmission electron microscopy(TEM). Clinical result shows the phenomena of conjunctiva hyperemia have lightened evidently three weeks after surgery and no inflammation in anterior chember. At the eighth week, filter blebs can be seen flat and dispersion exists in three eyes out of the six eyes in group A, while in two of the three eyes in group B, and these can seldom seen in control group. The pathology process is: control group is similar to experimental group. One to three weeks after surgery inflammation exists, then acute inflammation lightened and chronic inflammation increase, and disappear after eight weeks. The acute inflammation of group A is stronger than group B and control group, and lighten evidently three weeks after surgery, similar to the other two groups. At the twelfth week, the spaces in sclera of group A and B are remained , while scar formation is found in control group. The scar formation in group A and B is much lighter than control group, applied t test p<0.05, with notable difference. TEM shows at the eighth week, there are spaces in collagen fibers with a few fibroblasts aside in group A and B, while in control group collagen fibers arrange irregularly and fibroblastic activity is increased. We can thus draw conclusions that the amniotic membrane implant andSK GEL implant can inhibit the scar formation in the NPTS in rabbit eyes. That is to say, non-penetraling trabecular surgery with amniotic membrane and SK GEL implant is safe and effective and can inhibite scar formation.
