ObjectiveTo study the abnormality of multiple cytokine mRNA expression of bone marrow mesenchymal stem cells (BM-MSCs) from patients with multiple myeloma (MM), as well as to explore the role of these abnormalitis in the occurrence and development of MM. MethodsSemi-quantitiative RT-PCR was used to detect the mRNA expressions of IL-1β, IL-6. SCF, TPO, G-CSF in BM-MSCs from MM patients. ResultThe ratios of multiple cytokines mRNA level to β-actin mRNA (Rm) of BM-MSCs in MM patients were detected and were compared with those of normal controls and other hematological malignancies.The Rm ratios of IL-1β, IL-6, SCF and TPO in MM patinets were 0.6922 ±0.1646, 0.9221 ±0.1234, 0.7041 ±0.1109, 0.6770 ±0.1777, respectively.The Rm ratios of IL-1β, IL-6, SCF and TPO in other hematological malignancies were 0.1821 ±0.0593, 0.2371 ±0.0491, 0.3222±0.0436,0.5400±0.2511, respectively.The Rm ratios of IL-1β, IL-6, SCF and TPO in control group were 0.2849 ±0.1464, 0.4574 ± 0.0644, 0.4009 ± 0.0971, 0.3342 ± 0.1182, respectively.MM patients had higher mRNA expression levels of IL-1β and IL-6 than that of normal controls and other hematological malignanciesFor the expression of SCF, There is no significant differences between the transplanted patients and the normal controls ( P> 0.05 ), but we could see a the significant difference when the primary MM and the relapsed MM constrasted to the normal controls( P<0.05 ). The expression difference is significant between the MM patients and other hematological malignancies ( P<0.05 ) .For the expression of TPO, There is a significant difference between the MM patients and the normal controls (to the primary MM P<0.01, the other P<0.05 ) .When we saw a significant difference between the primary MM and other hematological malignancies( P<0.01 ), we didn't see the same when the relapsed MM and the auto-transplanted MM contrasted to other hematological malignancies .We could not find any G-CSF mRNA expression in all study groups within the sensitivity limitation of our methods.ConclusionIL-1β, IL-6, SCF and TPO are up-regulated at mRNA level in in vitro cultured BM-MSCs from MM patients and may play an important role in the disease pathogenesis.
