Objective:Construction of bone marrow stromal cells and myeloma cell fusion cells in vitro, and explore the biological characteristics of the fused cells.Methods : 1.Cultivation respectively between normal people and patients with multiple myeloma bone marrow mesenchymal Stem Cells(MSCs), and preliminary characterization of its biological characteristics.2. Cell tracer green fluorescent probe(CMFDA) and red fluorescent probe(CMTMR) individually tagged MSCs and myeloma cells through chemical promote melting agent polyethylene glycol(polyethylene glycol PEG- 1000) induced fusion, cell fusion model is set up.3. Fused cells biological characteristics research: Karyotype analysis, to determine whether fusion cell nucleus fusion; Identification of fusion cells dry-related genes C-myc, Sox2, Oct-4, etc expression and the expression of promote harmony correlation genes SIRP α and DC – STAMP.Results:1. Successfully isolated and cultured human bone marrow mesenchymal stem cells 2. Polyethylene glycol(PEG-1000) can mediate MSCs fused with myeloma cells 3. More than 80% of the number of chromosomes in the hybrid cell is about 90; Fused cells not only show the characteristic of BMSC, dry-related genes C- myc, Sox2, Oct- 4 express positive, but the fusion-related genes SIRP α and DC-STAMP also expressed positive.Conclusion:Between MSCs with myeloma cells can form a fused cell.Fusion cells have the potential to further a melt is one of the important reasons for the occurrence of multiple myeloma and development.
