PurposeThe key of clinical therapy on the open trauma and the skin injury was to close traumatic areas and establish barriers as soon as possible, but other treatments must be used when the loss of large area of skin was beyond the ability of self - repair. Artificial skin of tissue engineering provided a novel choice for all kinds of skin injuries. Many products about this had been used in patients and became commercial in other countries, but the institutes and companies were exploring this item in china in recent years. This study was to investigate the tissue characteristics and growth state of oral mucous fibroblasts (OMF) on the artificial scaffold of Chitosan - Collagen, to discuss feasibility about in vitro construction of artificial lamina propria by using artificial scaffold of Chitosan - Collagen and OMF, at last to provide the experiment basis for the preparation of o-ral mucous of composite Chitosan tissue engineering.MethodsThe Rat OMF was extracted and cultured in vitro. Under sterile condition, 2 x DMEM solution, serum of embryo cattle, collagen solution and Chitosan, or 2 x DMEM solution, serum of embryo cattle and collagen solution were mixed together into gel solution pro rata. OMFs were added into above gel solution to form oral mucous of artificial lamina propria. Exchange solution one time per day and watch the dynamical growth of OMFs and dynamical constriction of gel.
